This study aimed to examine the effects of exogenous substances such as glucose,salicylic acid and Ca2+ on eggplant(Solanum melongena L.)seeds germination at low temperature,and morphological and physiological changes of plug seedlings during storage and transportation.Eggplant cultivar ’Xiuniang’ of Chinese origin was used as experimental material.The study was carried out in two steps,at first step,seeds were soaked in three different concentrations of exogenous substances(glucose,salicylic acid and Ca2+),while soaking in water was used as a control treatment,and their effect on seed germination was evaluated.In second step,solutions of the said exogenous substances with different concentrations were sprayed on seedlings,water spray was used as control treatment,and morphological and physiological changes of plug seedlings during storage and transportation were evaluated.The findings of this study provide theoretical basis and technical support for eggplant seed germination and plug seedlings in winter and spring.The results of the study are as follows:1.Effects of seed soaking with exogenous substances on germination of eggplant seeds at low temperature.In low germination temperature condition(15℃)soaking of seeds in certain concentrations range of glucose,salicylic acid and Ca2+solution improve the germination rate,germination potential and germination index of eggplant seeds,and promote the growth of hypocotyls and radicles.Among the treated concentrations(2~24 mmol·L-1 glucose,5~25 mmol·L-1 Ca2+,and 0.1~1.6 mmol·L-1 salicylic acid)6 mmol·L-1 glucose,20 mmol·L-1 Ca2+,0.4 mmol·L-1 salicylic acid improved seed germination as well as hypocotyls and radicles growth significantly.The results revealed that soaking of eggplant seeds with Ca2+ was effective than glucose and salicylic acid as well as from control treatment.2.Effects of exogenous substances on quality of shipping and the ability to restore growth after transplanting of seedlingsThe quality of shipping of eggplant plug seedlings decreased significantly with the extend of shipping time in control treatment(CK).Spraying with exogenous substances increased the seedling index,root cap ratio,and specific leaf weight.Also the growth rate of the central leaves in eggplant plug-seedlings were increased.The optimal concentration was 4.8 mmol·L-1 glucose,12 mmol·L-1 Ca2+,2.4 mmol·L-1 salicylic acid.The results revealed that spaying eggplant seedlings with glucose was effective than salicylic acid and Ca2+,as well as from control treatment.3.Effects of exogenous substance on related metabolism index of storage and transportationThe elated metabolism index of shipping of eggplant plug seedlings without treatment decreased significantly with the extend of shipping time.The exogenous glucose,Ca2+ and salicylic acid spraying had a positive effect on the accumulation of chlorophyll content,soluble sugar content and soluble protein content in the plug seedlings during storage and transportation.So that the material metabolism has always been maintained at a relatively stable level.The effect of salicylic acid and glucose was better than Ca2+4.Effects of exogenous substance on good shipping quality of eggplant plug seedlingsThe content of active oxygen species(ROS),malondialdehyde(MDA)and the increasing trend of relative conductivity in the leaves of eggplant seedling were significantly decreased by different concentrations of treatment of glucose,salicylic acid and Ca2+.And the exogenous substance treatment also promote the activities of superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT)and the accumulation of proline content.So that the degree of cell osmotic potential and plasma membrane peroxidation to maintain a relatively stable level,which enhance the ability of plants to remove reactive oxygen species,improve the good shipping quality of eggplant plug seedlings.The order of good shipping quality of different exogenous materials is glucose>salicylic acid>Ca2+.It can be concluded that the three kinds of exogenous substances have a positive effect on improving the low temperature germination effect of eggplant seeds and promoting the good shipping quality and quality of shipping with the comprehensive analysis of the seed germination index,changes of morphological indexes and physiological indexes in plug seedlings during storage and transportation and the ability to restore growth after planting of seedling.Which shows that the soaking effect of 6 mmol·L-1 glucose,20 mmol·L-1 Ca 2+and 0.4 mmol·L-1 salicylic acid were better.And the spraying effect of 4.8 mmol·L-1 glucose,12 mmol·L-1 Ca2+,2.4 mmol·L-1 salicylic acid is better.
