Objective: In order to determine the chemical constituents and pharmacological activities of Iodes vitiginea,the chemical constituents of ethyl acetate extract and nbutanol extract were studied,and the pharmacological activities of extractum from different polar parts of Iodes vitiginea were screened.Methods: In this paper,thin layer chromatography,silica gel column chromatography,high performance liquid chromatography,D101 macroporous resin column chromatography,reversed-phase ODS column chromatography,Sephadex LH-20(hydroxypropyl Glucan gel)column chromatography,semi-preparative liquid chromatography and other methods were used to separate and purify the compounds from the root of Iodes vitiginea.The structures of the isolated compounds were identified by modern spectral analysis techniques.CCK8 method was used to screen the cell activity of extracts from different polar parts of Iodes vitiginea.Results:(1)Twelve compounds were isolated and identified from ethyl acetate extract.Two compounds were isolated from n-butanol and identified.These compounds are(S)-5-ethyl-8,8-dimethylnonanal(1),3β-O-vanilloyl-taraxerol(2),3β-formyl-taraxeryl(3),β-sitosterol(4),cyclo(L-pro-L-Leu)(5),(R)-2,3-Dihydroxypropyl-stearate(6),3α,11β-dihydroxy-pimara-8(14),15-diene(7),2′,3′-dihydroxy-propyl-pentadecanoate(8),β-sitosteryl-3-O-β-D-glucopyranoside-2′-O-palmitate(9),(6’-O-palmitoyl)-sitosterol-3-O-β-D-glucoside(10),palmityl-linolacyl-glucoside(11),β-carotenoside(12),pinoresinol-4’-O-β-D-glucopyranosyl(1→2)-β-D-glucopyranoside(13),resinin-4′-O-β-D-glucopyrano-4"-O-β-D-furapigenin(14).(2)CCK8 method was used to study the antitumor activity of extractum from different polar parts of Iodes vitiginea.Rat glioma cells were used as receptor cells.The results showed that the n-butanol extract had inhibitory effect on cell proliferation,and the inhibitory effect was the best when the drug concentration was 800 mg/L,with a certain dose dependence.Conclusion: A total of 14 compounds were isolated and identified,all of which were isolated from this plant for the first time.N-butanol extract showed an inhibitory effect on rat glioma cells,and the inhibitory effect was better when the concentration was 800mg/L,with a certain dose dependence.
