Purpose:1.The effect of stilbene glucoside on postmenopausal osteoporosis;2.To observe the effect of stilbene glucoside on estrogen,calcium and phosphorus metabolism in the treatment of postmenopausal osteoporosis;3.To further explore whether stilbene glucoside plays an anti osteoporosis role through KL /FGF23 signaling pathway.Materials and methods:Forty 3-month-old female SD rats were selected and divided into 4 groups with 10 rats in each group.Including: normal group,model group,experimental group(stilbene glycoside group),control group(raloxifene group).Except the normal group,the other groups were treated with bilateral ovariectomy to establish PMOP rat model.The drug was administered for 8 weeks from the 7th day after operation.After the last administration,the bone mineral density of anesthetized rats was detected by dual energy X-ray,take out the material then conduct the vertebral bone stress was detected,the blood calcium and phosphorus were detected by automatic biochemical analyzer,the estrogen and FGF23 were detected by ELISA,the morphology of the right femur was observed by HE staining,and the KL protein expression level was detected by immunohistochemistry.Result:1.Stilbene glucoside can effectively treat postmenopausal osteoporosis,improve bone structure and restore bone stress: compared with the normal group,the BMD of the model group was significantly lower,the difference was statistically significant(P<0.01);Compared with the model group,the BMD of the experimental group and the control group increased,and the difference was statistically significant(P<0.05);After the establishment of osteoporosis model,the body weight of rats in the model group was significantly higher than that in the normal group(P<0.01);After treatment,the weight of the experimental group and the control group decreased significantly,and the difference was statistically significant compared with the model group(P<0.05);Compared with the normal group,the uterine index of the model group was lower,and the difference was statistically significant(P<0.01);Compared with the model group,the uterine index of the experimental group and the control group was higher,and the difference was statistically significant(P<0.05);He staining showed that the trabecular structure of normal group was complete and continuous,and the proportion of trabecular bone was high;In the model group,the bone trabeculae were sparse,the gap became larger,and the continuity became worse;The trabeculae of the experimental group and the control group were dense,continuous and had more trabeculae;Compared with the normal group,the bone stress of the model group decreased significantly(P<0.05);Compared with the model group,the bone stress indexes of the experimental group and the control group were increased,and the difference was statistically significant(P<0.05).2.Stilbene glucoside can regulate the content of estrogen and blood calcium and phosphorus in ovariectomized rats,and affect bone metabolism: compared with the normal group,the content of estrogen in the model group was lower,and the difference was statistically significant(P<0.01);Compared with the model group,the estrogen levels of the experimental group and the control group were improved in different degrees,and the difference was statistically significant(P<0.05);The serum calcium content of model group was lower than that of normal group(P<0.01);Compared with the model group,the serum calcium content of the experimental group and the control group increased,the difference was statistically significant(P<0.05);The content of serum phosphorus in the model group was higher than that in the normal group(P<0.01);Compared with the model group,the serum phosphorus content of the experimental group and the control group decreased,the difference was statistically significant(P<0.05);3.Stilbene glucoside could affect the protein expression of KL and FGF23: compared with the normal group,the content of FGF23 in the model group was higher,and the difference was statistically significant(P<0.01);Compared with the model group,the content of FGF23 in the experimental group and the control group decreased,and the difference was statistically significant(P<0.05);The expression of KL protein in the model group was lower than that in the control group;Conclusion:1.Stilbene glucoside can effectively affect postmenopausal osteoporosis,promote bone formation and restore bone structure;2.Stilbene glucoside can up regulate estrogen level,reduce calcium loss,restore calcium and phosphorus regulation,and affect bone metabolism in ovariectomized rats;3.Stilbene glucoside can promote the expression of KL protein,inhibit the expression of FGF23 protein,and play the role of anti osteoporosis,but whether kl-fgf23 signaling pathway plays a role needs to be further verified.
