Objective: To observe the effect of Huanglian Wendan Decoction on KM male mice with type 2 diabetes(T2DM)successfully modeled by gavage with traditional Chinese medicine,and to preliminarily explore the effect of Huanglian Wendan Decoction on the browning of white adipose tissue in mice,based on PI3K/AKT Pathway to improve lipid glucose metabolism and insulin resistance in T2 DM mice;to explore the prevention and treatment effects of Huanglian Wendan Decoction on T2 DM and related mechanisms from the perspective of white adipose tissue browning;to further study the intervention of Huanglian Wendan Decoction on metabolic diseases and The mechanism of action provides experimental data support.Methods: Choose healthy 4-week-old male KM mice,fed with high-and high-sugar feed combined with small-dose intraperitoneal injection of streptozotocin(STZ)to induce the establishment of a T2 DM mice model.After modeling,the mice were divided into blank groups(KD group),The control group(KK group)and the traditional Chinese medicine group(KY group),to investigate the effects of Huanglian Wendan Decoction on fasting blood glucose(FBG),glucose tolerance(IPGTT),C-peptide,and body weight in mice,and observe the effect of the prescription on the browning of subcutaneous inguinal white adipose tissue(i WAT)in mice and The influence of related protein PI3 K,AKT,PGC-1α,UCP-1 expression.result:1.FBG and weight related results: After intervention,the FBG of the control group was significantly higher than that of the blank group(P<0.001);the FBG of the traditional Chinese medicine group was significantly lower than that of the control group(P<0.001).In terms of body weight,at the 9th week(before administration)and13th week(after administration),compared with the blank group,the body weight of the mice in the control group increased significantly(P<0.001);at the 13 th week(administration)After),compared with the control group,the traditional Chinese medicine group increased significantly(P<0.001);the weight change,compared with the control group,the traditional Chinese medicine group increased significantly(P<0.01).2.IPGTT: Compared with the blank group,the area under the blood glucose-time curve(AUC)of the pre-administration group and the post-administration control group after model building was extremely significantly increased(P<0.01);The AUC of the traditional Chinese medicine group was significantly reduced after treatment(P<0.01).3.Western Blotting detected the expression of PGC-1α and UCP-1 in i WAT:Compared with the blank group,the expression of PGC-1α in the Chinese medicine group increased significantly(P <0.01);compared with the control group,the PGC-1α expression in the Chinese medicine group also increased significantly(P <0.01).In terms of the expression of UCP-1 protein,the expression of the traditional Chinese medicine group was significantly higher than that of the blank group and the control group(P <0.01),and there was no difference between the blank group and the control group.4.Western Blotting detected the expression of PI3K/AKT pathway in i WAT:Compared with the blank group,the decrease of PI3 K expression in adipose tissue in the Chinese medicine group was statistically significant(P<0.05),while the expression of AKT decreased significantly(P<0.01);the Chinese medicine group The expression levels of the two proteins were significantly higher than those of the control group(P <0.01).5.Mice i WAT histology HE staining: In the traditional Chinese medicine group,droplets were found to be smaller,accompanied by a small number of small lipid droplets around the local large lipid droplets.Compared with the control group,the volume of lipid droplets was significantly reduced,the cytoplasm was deeply stained,and the capillaries increased.The appearance of similar brown adipocytes is a manifestation of the browning of white adipose tissue.in conclusion:1.In this experiment,Huanglian Wendan Decoction can significantly improve the weight loss of T2 DM mice,reduce FBG,reduce AUC,etc.,and achieve the effect of reducing insulin resistance.2.In this experiment,Huanglian Wendan relative expression of PI3K/AKT in the KD group was decreased compared with KK.The intervention of Huanglian Wendan Decoction made the expression of PI3K/AKT pathway in the KY group significantly higher than that in the KD group,but it still did not return to the normal level.4.In this experiment,compared with the KD group,the changes in the expression of PI3K/AKT pathways in the KK and KY groups are different;while the differences in the expression of proteins such as PGC-1α and UCP-1 are mainly reflected in the KK group and the KY group between.This shows that the PI3K/AKT pathway is more sensitive to changes in the levels of FBG and insulin resistance in the body.
