Objective: To observe the effects of electroacupuncture at the "Siguan" acupoints and their corresponding single points on the PSD rats,by the test index of brain-derived neurotrophic factor(BDNF),Kinase family B(Tropomyosin-related kinase B,Trk B)protein expression and behavioral changes,to study the antidepressant mechanism of electroacupuncture at Siguan acupoints in the treatment of PSD,and to explore whether Hegu acupoints and Tai chong acupoints have synergistic effectwhen they are used to treat PSD rats.Methods: Forty SD rats were randomly divided into blank group,model group,Siguan group,Hegu group and Taichong group,with 8rats in each group.Except for the blank group,all rats were modeled with PSD.except for the blank group and model group were not treated with acupuncture,other groups were treated with electroacupuncture at corresponding points for 3 weeks.The depressive behavior changes of rats were observed by open-box experiment(number of grids passing,number of erection modification times)and sugar water consumption experiment before modeling,1,7,14,and 21 days after modeling.The content of BDNF and Trk B in the hippocampus will be determined after treatment.Results:1.After PSD modelling,the number of grids in the open-box,erection modifications the ratio of sugar-water consumption and other behaviors in each group were significantly lower than those of the blank group(P<0.01).2.From the perspective of the number of grids in the open-box experiment,the three groups of acupuncture can significantly increase the number of grids of rats(P < 0.05);among them,the Hegu group increased significantly in the 14-21 days(P < 0.01),Taichong The improvement was significant in the first 7 days of the group(P<0.01),and the Siguan group improved significantly throughout the whole process(P<0.01);on the 21 st day,the Siguan group increased significantly than the Hegu and Taichong groups(P<0.01),there was no significant difference between Hegu group and Taichong group(P>0.05).3.From the perspective of the number of erection modifications in the open-box experiment,the three groups of acupuncture can significantly increase the number of erection modifications in rats(P<0.05);among them,the Hegu group improved significantly in the first 7-14 days(P < 0.01),While the Siguan group and Taichong group improved significantly in the whole process(P<0.01);on the21 st day,the Siguan acupoint group improved more significantly than the Hegu group and the Taichong group(P<0.01),and the Hegu group and the Taichong group There was no significant difference between them(P>0.05).4.From the perspective of sugar-water consumption ratio,the three groups of acupuncture can significantly increase the sugar-water consumption ratio of rats(P < 0.01);the three groups increased significantly in the whole process(P < 0.01);on the 21 st day,the Siguan point group was compared The frequency of Hegu group and Taichong group increased more significantly(P < 0.01),and the increase of Taichong group was more significant than that of Hegu group(P < 0.01).5.After the treatment,electroacupuncture at Siguan,Hegu,and Taichong points can significantly increase the number of BDNF and Trk B positive cells in PSD model rats(P < 0.01).Among them,electroacupuncture at Siguan and Taichong points In terms of increasing the number of BDNF and Trk B positive cells,they were better than electroacupuncture at Hegu(P < 0.05).Conclusion:.Electroacupuncture at Siguan points can significantly improve the behavioral indicators of PSD rats.When increasing the number of grids in the open-box experiment,Hegu and Taichong have a synergistic effect,and increase the number of erection modifications and the consumption of sugar and water in the open-box experiment.During the comparison,the Taichong point plays a major role;its mechanism of action may be related to increasing the expression of BDNF and Trk B protein,thereby improving neuron survival,development and function;the synergistic effect of Siguan points in the treatment of PSD is partially reflected,but it still needs further research.
