Effects Of Shenfu Injection On The Expressions Of HIF-1α And VEGF-A In Neonatal Rats With Hypoxic-ischemic Brain Injury

Objective:This study was to investigate whether Shenfu injection has therapeutic effect on hypoxic ischemic brain injury in neonatal rats,and whether this effect is related to the expression of HIF-1α and VEGF-A.Methods:A total of 120 SPF 7-day-old neonatal rats were selected,and 30 of them were randomly assigned to blank control group(group B)without any intervention.In addition,30 rats were randomly selected as the sham operation group(group K).During the operation,the left common carotid artery was dissected and the wound was sutured,and the rats were fed and breathing air normally.The remaining 60 newborn rats with hypoxic-ischemic brain injury model were made according to the classical RICE method.After modeling,the neurologic loss behavioral score was used for judgment.Those who met the requirements were included in the experiment and divided into model group(group M)and Shenfu injection group(group Z)according to the random number table method.Then,according to different observation time points,the four groups of neonatal rats were divided into five subgroups: 6h,12 h,24h,72 h and 168 h,with 6rats in each group.Model group(group M)was injected intraperitoneally with normal saline at a dose of 20ml/kg·day after hypoxia in anoxic device,while Shenfu injection treatment group(group Z)was injected intraperitoneally with Shenfu injection at a dose of 20ml/kg·day.The rats were sacrificed according to the corresponding observation time points,and the brain water content was calculated by weighing the brain.The changes in the expression of HIF-1α and VEGF-Am RNA in the cerebral cortex were detected by fluorescence quantitative PCR method,and then statistically analyzed.The pathological morphology of the brain tissues of each group was observed under light microscope by HE staining and Nissl staining.Results:1.Compared with the blank control group and the sham operation group,the model group and the Shenfu injection treatment group showed different degrees of neurological function loss after hypoxia(P<0.05),and the phenomenon of turning over and clipping tail left rotation,etc.2.The pathological sections of the brain tissue after the model were changed to varying degrees,such as the loose arrangement of nerve cells and the change of the number of Nissellite,etc.,while the observation of the pathological sections of the Shenfu injection group showed that the injury degree of nerve cells was less;3.Compared with the blank control group and sham operation group,the brain moisture content of the model group and Shenfu injection treatment group increased,and the brain moisture content of Shenfu injection after treatment decreased compared with the model group;4.HIF-1 α m RNA in blank control group,and traces of expression in the control group,model group and the cords attached injection treatment group were increased in the control group(P<0.05),and the same time trend,began a large number of expression,in 6h,12 h,24h is peak,and then decreased 72 h,168h expression,and attached injection treatment group expression quantity in each time point compared with model group increased(P<0.05);5.VEGF-A m RNA expression in A blank control group and the control group,model group and the cords attached injection treatment group were increased in the control group(P<0.05),and the same time trend,began an large number of expression,in 6h,12 h,24h is peak,and then decreased 72 h,168h expression,and attached injection treatment group expression quantity in each time point compared with model group increased(P<0.05).Conclusion:1.Shenfu injection treatment group has significantly reduced pathological damage compared with model group;2.Shenfu injection treatment group may reduce the symptoms of cerebral edema in the acute stage of brain injury;3.Shenfu injection has a protective effect on the brain in the early stage of HIE,which may By regulating the HIF-1α/VEGF-A pathway,it is time-dependent.
