Objective:To observe the effects of Rougan Tongluo Decoction on the degree of neurological deficit,cerebral infarction area and cell damage in rats with cerebral ischemia-re perfusion injury,and to explore the effect of Rougan Tongluo Decoction on cerebral ischemia reperfusion injury from the perspective of autophagy mechanism.Methods:120 SD rats were randomly divided into 4 groups:blank group,sham operation group,model group and Rougan Tongluo Decoction group.The blank group was not treated,the sham operation group and the model group were given distilled water gavage,and the Rougan Tongluo Decoction group was gavaged with high-dose Rougan Tongluo decoction,all of which were pretreated for one week.30 minutes after the last gavage,a cerebral ischemia-reperfusion model was established,and the brain was taken at 4 time points of 3、6、12 and 24 hours of reperfusion.The front of the brain was taken to score the neurological deficit of the rat,and after taking the brain,we performed TTC staining to observe the area of cerebral infarction,HE staining to observe the patho morphological changes of ischemic tissue,and immunohis to chemical method to detect the expression of Beclin 1 and LC3 protein.Results:Compared with the sham operation group,the neurological deficit score of the model group increased(p<0.01),the cerebral infarction area increased(p<0.01),the neuronal cell damage on the infarcted side was aggravated,and the expression of Beclin 1 and LC3 increased(p<0.01).Compared with the model group,the neurological deficit scores in the Rougan Tongluo Decoction group decreased(p<0.05),the cerebral infarction area of rats was reduced(p<0.01),the neuronal cell damage on the infarct side was significantly reduced,and the expressions of Beclinl and LC3 were reduced(p<0.01).Conclusion:Rougan Tongluo Decoction can down-regulate the expression of autophagy proteins Beclinl and LC3 after ischemia-reperfusion injury,inhibit cell autophagy,thereby reducing cerebral ischemia-reperfusion injury.
