Effects Of Zhiyan Mixture On The Levels Of IL-5 And LTB4 In Mycoplasma Pneumoniae Infection Model Rats

Objective:To analyze the effects of Zhiyan Mixture on IL-5 and LTB4 levels in rats infected with Mycoplasma pneumoniae through animal experiments,and to preliminarily explore the therapeutic mechanism of Zhiyan Mixture,in order to provide a basis for clinical application.Methods:48 SPF Wistar rats,male and female,with body weight of 200±20g,were randomly divided into blank group,Mycoplasma pneumoniae model group(MP group for short),azithromycin group,and chiyan mixture group,with 12 rats in each group.The rat model of Mycoplasma pneumoniae was constructed by repeated nasal infection of MP after being fed separately in cages for one week.The blank group was intranasally injected with equal volume of normal saline,and the other groups were intranasally inoculated with 100ul MP bacteria solution(concentration 107 CCU/ml),once a day for consecutive 3 days.The four groups were given continuous administration for 10 days.The azithromycin group was given 5 days,and the same amount of normal saline was given intragastric administration for the next 5 days.Serum samples were taken from the abdominal aorta of rats and complete lung tissues were extracted from each group at 5 and 10 days after administration.The levels of IL-5 and LTB4 were determined by enzyme-linked immunosorbent assay(ELISA).Pathological observation was made by HE staining after paraffin embedding section of lung tissue.Results:1.Comparison of body weight and lung wet weight index of rats:on day 5,compared with blank group,lung wet weight index of MP group,azithromycin group and Zhiyan mixture group was significantly increased(all P<0.05),while body weight was significantly decreased(all P<0.05);Compared with MP group,the body weight of the remaining three groups was significantly increased(all P<0.05),and the lung wet weight index was significantly decreased(all P<0.05).There were no significant differences in body weight and lung wet weight index between azithromycin group and Zhiyan mixture group(all P BBB 0 0.05).On the 10th day of observation,there were no significant differences in body weight and wet lung weight index(all P BBB 0 0.05),except that the body weight of blank group was significantly increased(P<0.05).2.ELISA results:Compared with the blank group on the 5th day,the contents of IL-5 and LTB-4 in MP group were significantly increased(all P<0.05);Compared with MP group,the contents of IL-5 and LTB-4 in azithromycin group and Zhiyan mixture group were significantly decreased(all P<0.05).Compared with azithromycin group,there was no significant difference in the content of Zhiyan mixture group and azithromycin group(P BBB 0 0.05 for all),but it was significantly higher than that of blank group(P<0.05 for all).On the 10th day of observation,there was no significant difference in the level of IL-5 among the four groups(all P>0.05),but there was significant difference in the level of LTB-4 between the azithromycin group and the Zhiyan mixture group(all P<0.05).3.HE staining results:the alveolar wall of blank group was not significantly thickened,the structure was basically intact,and there was very little inflammatory cell infiltration;in MP group,large area of alveolar wall moderate thickening was observed,accompanied by a large amount of inflammatory cell infiltration,local bronchus and a large amount of peripheral lymphocyte infiltration,peripheral lymphocyte infiltration ring,a large amount of blood vessels congestion was observed,and red blood cells were observed in the alveolar cavity.In both the azithromycin group and the Zhiyan mixture group,lung tissue structure was damaged,with mild local bleeding,moderate alveolar wall thickening,partial alveolar wall thickening and rupture,and less inflammatory cell infiltration.There was no significant difference between the groups on day 10 and day5.4.Lung histopathological score:on the 5th day,compared with the blank group,the lung histopathological score of MP group,azithromycin group and Zhiyan mixture group was significantly higher(all P<0.05);Compared with MP group,lung histopathological scores in azithromycin group and Zhiyan mixture group were significantly lower(both P<0.05).There was no significant difference between azithromycin group and Zhiyan mixture group(P BBB 0.05).On the 10th day of observation,there was no significant difference among groups(all P>0.05).Conclusion:1.Zhiyan Mixture can improve the clinical symptoms of MP-infected rats and reduce the inflammatory response of their lung tissue.2.The levels of IL-5 and LTB-4 in serum of MP infected rats were significantly increased,and Zhiyan Mixture could significantly reduce the levels of IL-5 and LTB-4.3.Zhiyan Mixture may play a certain therapeutic role in the treatment of MP infected patients by regulating some cytokines to reduce the levels of IL-5 and LTB-4.
