In the treatment of gynecological diseases,antibiotics are the main drugs in the market,and the packing lactic acid bacteria as the way of drug treatment,all of them have their own drawbacks.Related studies have found that oral Lactobacillus microecological preparation has the same curative effect on gynecological diseases,but it is doubtful whether it can enter the reproductive system after its entry into the digestive system.Specifically,oral lactobacillus is directly into the reproductive tract,or indirectly affects the vaginal environment.To investigate this question,we introduced g Wiz-GFP plasmid Gwiz into Lactobacillus saliva in the form of electrical transformation,which was designed for high level expression of genes in a wide range of mammalian cells and could be directly used in vivo and in vitro without endotoxin.GWiz-GFP Lactobacillus saliva was constructed as the solution for gastric perfusion.Forty female SPF rats were divided into four groups:healthy PBSs(groupⅠ),model PBSs(groupⅡ),model bacteria solution(groupⅢ)and healthy bacteria solution (groupⅣ).GroupⅢandⅡwere injected with estradiol phthalate to make their pseudoestrus,and then the pathogenic bacteria composed of Escherichia coli and Staphylococcus aureus at 1:1 were artificially infected with external vagina to construct the model mice.The feces and the green fluorescence expression of vaginal mucosa of four groups of mice were measured by g Wiz-GFP Lactobacillus saliva daily intragastric administration of PBS solution of the same dose,control groupⅠand control groupⅡfor 20 days.The changes of the number of two major pathogens in the vagina and the distribution of bacteria in vaginal and intestinal epithelium and the migration of green fluorescence in vivo were recorded by detecting the lactobacillus g Wiz-GFP in vagina.The results showed that the number of vaginal lactic acid bacteria increased from 103CFU/g to 105 CFU/g,Escherichia coli and Staphylococcus aureus decreased to about 10~2CFU/g(P<0.01),10~4CFU/g(P<0.05),and the vaginal microflora was adjusted well.The PH of model mice recovered from about 8.2 to that of H 2O2 in acidic environment near 6.4 from 1.5mmol/L to 2.73mmol/L(P<0.01),and the vaginal environment of model mice recovered well.The stable expression of green fluorescence in vaginal mucosa and their upper epidermis proved that oral lactobacillus g Wiz-GFP could enter the large intestine and be colonized in the genital tract,which could adjust the environmental microecological balance,achieve the treatment and prevent bacterial vaginitis. |
