Licorice is rich in aboveground resource,it has high content of total flavonoids,and it has a variety of pharmacological activity such as anti-inflammatory,anti-oxidation and so on,so the aerial part of licorice has a very high value in use.People use the leaves of licorice as tea for treatment of CNP and got good results.However,due to the lack of in-depth and more systematic theoretical study,aboveground resources of licorice has not been effectively developed and utilized at this stage,only as a pasture in pastoral use or discarded,resulting in a waste of natural resources.For better development and utilization of licorice resources,this paper studies the fat-soluble constituents in aerial parts of licorice.By purification of constituents,and qualitatively analyzed by HPLC-MS,examining the main constituents chromatography method,and do the antioxidant and anti-CNP research activity.The results are as follows:(1)The establishment of HPLC fingerprint and qualitative analysis by HPLC-MS detection for fat-soluble constituents in aerial part of licorice.Establishing HPLC fingerprint of the aerial parts of the fat-soluble constituents of licorice,HPLC fingerprint chromatographic conditions:DIKMA Diamonsil(?)-C18 analytical column(4.6×250mm,5μm),acetonitrile-0.1%phosphoric acid-water gradient,the volumetric flow rate is 1mL/min,column temperature is 35℃,detection wavelength is 190nm-400nm.Results shows that HPLC-DAD fingerprint method can show fat-soluble constituents in aerial part of licorice comprehensively.Four kinds of constituents was determined by HPLC-MS,named quercetin,genistein,pinocembrin and uralenin.This study make establishment for the constituents study of upground part of licorice.(2)Study of enrichment and purification methods of total fat-soluble constituents in aerial parts of licorice.To find an effective method of enrichment of the aerial parts of fatsoluble constituents of licorice and to explore and optimize process conditions.Solution samples was adsorbed in resin A and was washed with purify water,then loaded on the top of macroporous resin,eluting with ethanol.Results shows that SZ-1 resin combined with resin A could effectively enrich the total constituents.The preferred condition for the procedure was as following:the weight of resin A was equal to samples,SZ-1 resin was 10 times of the samples’weight.The volume of 70%ethanol for elution was 6 times of the column volume.The purity of total constituents reached to(51.6±2.0)%and the recovery rate was(73.8±3.5)%.This method can solve the sample problem and is convenient,which has a good application foreground in flavonoid enrichment in licorice.(3)Verify that the aerial parts of licorice has good antioxidant activity.Aboveground portion of licorice is extracted into chloroform and ethyl acetate extracts by solvent extraction system.Samples was determined by DPPH radical,hydroxyl free radicals,reducing power and total antioxidant capacity.Results shows that total fat-soluble constituents in aboveground portion of licorice and each segment has strong antioxidant capacity,which the ethyl acetate extract is the strongest.After enrichment,the various parts of the antioxidant capacity was increased.The fat-soluble constituents in aboveground licorice have good antioxidant capacity,it is developing in natural antioxidants development.(4)Licorice upground fat-souble constituents has good anti-CNP activity.Chemically induced inflammation in rats to establish the model of CNP,studying the efficacy of anti-CNP activity of enriched fat-souble constituents in aerial part of licorice.QianLiekang as a positive control,compared to PAPL.The CNP model was established successfully,the results shows that PAPL has a certain degree of anti-CNP activity.Results of pathological analysis indecated stronger activity in the part of PAPL than D APL.
