Study On The Anti-inflammatory And Antibacterial Efficacy Parts Of Jinlianhua Decoction

Jinlianhua decoction is composed of the flowers of Trollius chinesis,the flowers of Taraxacum mongolicum,the roots of Pueraria lobata,the leaves of Isatis indigotica,and the leaves of Perilla frutescens.It is one of the descriptions which were published by Chinese health authority for the prevention of atypical pneumonia(SARS).It can clear away heat and toxic material,and resolve the exterior and dissipate cold,and is indicated for common cold,pneumonia,cough,acute inflammation,and influenza.In order to efficiently develop this description,we extracted this description with boiling water according to the traditional usage of Chinese medicine,and carried out the investigations of human intestinal bacterial transformation and intestinal absorption,and then performed the effective part screening in combination with the antibacterial and anti-inflammatory screen.During the effective part screening,we first used the macroporous resin column chromatography to obtain 20,40,60,80,and 95%ethanol eluting part,then conducted the content determination of the major bioactive compounds,and finally,performed the antibacterial and anti-inflammatory study.The results showed that most of the compounds in the extract of Jinlianhua decoction can be absorbed by the human body after transformed by intestinal bacteria,the yield of 40%and 60%ethanol eluting parts were the two highest ones after separated by the macroporous resin column charomatography,and the contents of the major bioactive compounds such as puerarin,orientin,chlorogenic acid,and rosmarinci acid were also higher in both parts.Although the antibacterial and anti-inflammatory activities of the 40 and 60%ethanol eluting parts were not the best,they were more potent than the crude extract.In spite of the better bioactivities of other eluting parts,their yields were far less than those of the 40 and 60%ethanol eluting parts.In conculsion,we consider that the bioactive compounds of Jinlianhua decoction can be enriched by macroporous resin column charomatography eluted with water followed by high concentration ethonal such as 80%and 95%ethanol during the further drug research and development study...
