| Objective In the medical treatment of colorectal cancer,platinum-based drugs are played the most important role in no matter which chemotherapy projects.Nevertheless,platinum-based drugs are easy to drug resistance,it limited the choice of chemotherapy projects by doctors.Plasma Activity Media(PAM)is a kind of solution which is produced by treating the liquid medium with cold atmospheric plasma(CAP).A lot of studies have shown that PAM could inhibit tumor cells.In this study,the plasma generator was used to treat DMEM with helium(He)as the working gas to prepare PAM,and research the effects and related mechanisms of PAM on human colorectal cancer cells,and provide new ideas and methods for clinical treatment of colorectal cancer.Method This study includes:the study of cells’ viability,migration,invasion,and its related mechanism.First of all,the natural drug-resistant or artificial drug-resistant colorectal cancer cell lines were treated with PAM.Then,Cytotoxic drugs were used to study whether this cell line pretreated with PAM were more sensitive to these drugs.And we use wounder-healing test and Transwell test to detect the migration and invasion.In terms of mechanism,Westernblot was used to detect the expression of various proteins,including ERK,p38,GS3Kβ,β-catenin,c-myc and ITGB4,after PAM treatment to explore the effects of PAM on cell pathway.Result 1.Cell viability test implies that PAM has obvious inhibition with HT-29 cell line(P< 0.001),and this inhibition was time-dependent and dose – dependent.Cytotoxic drugs were added after pretreatment with PAM.Compared with PAM alone and chemotherapy alone,the cell viability of the combined treatment group was more significantly reduced,and the above conclusion was still valid whether it’s in some resistant cell line or not.2.In migration and invasion test,PAM group and combined treatment group could inhibit the migration and invasion ability of HT-29 cells,and the combined treatment group had a better inhibition efficiency.3.At the same time,Westernblot results showed that PAM could inhibit the expression of c-myc with its related proteins upstream and downstream,thus inhibiting the ability of tumor cells to metastasize to distant regions.Conclusion PAM‘s effects,which were single use or in combination with cytotoxic drugs including cell viability inhibitory,increasing the sensitivity of drug-resistant cells,migration and invasion,on human colorectal cancer cell lines is obvious.This inhibition of the proliferation and metastasis is achieved by inhibiting the expression of c-myc and its upstream and downstream. |
