Background In recent years,the incidence of gastric cancer has continued to rise,especially in developing countries,such as my country.With more and more research reports on this malignant tumor of the digestive system,people pay more attention to it.After the founding of the People’s Republic of China,China’s economic level has grown rapidly,the people’s diet and living standards have been greatly improved compared with the past,and the disease spectrum has also changed quietly.However,this improvement of living standards did not reduce the incidence of gastric cancer,but caused more people to suffer from gastric cancer due to the disordered diet.In recent years,the number of new cases of gastric cancer has greatly increased,and due to the lack of obvious clinical symptoms in the early stage,the initial diagnosis of patients is often in the middle and late stages.The clinical treatment methods corresponding to this stage are relatively simple,and the current popular immunotherapy has little effect,the prediction of the patient’s treatment prognosis depends only on the TNM stage of the tumor.Therefore,in view of the current situation of gastric cancer in my country,it is imperative to find potential targets for immunotherapy and biomarkers that can effectively predict the prognosis of patients.There is an inseparable relationship between the occurrence and development of gastric cancer and the tumor microenvironment(TME).Among them,tumor-associated macrophages(TAMs)and tumor-associated neutrophils(TANs)are important cellular components involved in tumor-related inflammation,and play an important role in tumor cell migration,invasion,and metastasis.play the most critical role in progress.To clarify the infiltration of TAMs and TANs in gastric cancer and their relationship with clinicopathological features and prognosis can provide new research ideas for the immunotherapy and prognosis analysis of gastric cancer.Aims To observe the infiltration of TAMs and TANs in gastric cancer tissue,and analyze their relationship with clinicopathological features and prognosis of gastric cancer patients.Method The TCGA database was used to analyze the infiltration of TAMs and TANs in gastric cancer tissue and whether it was related to the prognosis of patients.Then,89 cases of paraffin-embedded gastric cancer tissue samples were collected,and HE staining was used to observe the infiltration of TANs in gastric cancer tissue.The infiltration of TAMs in gastric cancer tissue was observed by chemical method,and its correlation with the clinicopathological characteristics and prognosis of patients was analyzed by statistical methods.Result(1)The infiltration of TAMs and TANs in 785 gastric cancer tissues was searched through the TCGA database,and the median number of TAMs and TANs in these cases was divided into high and low infiltration,and then correlated with the 5-year survival of gastric cancer patients in this sample.rate is analyzed.It was found that the 5-year survival rate of patients with high TAMs infiltration was lower than that of patients with low infiltration(P<0.05),while there was no significant difference in the 5-year survival rate between patients with high TANs infiltration and low infiltration(P>0.05);(2)The results of immunohistochemical staining showed that the infiltration ofTAMs was significantly correlated with vascular infiltration(P<0.001),and was correlated with lymph node metastasis and TNM stage(P<0.005),but there was no significant correlation with the gender and age of patients.sex(all P>0.05).The 5-year survival rate of patients with high TAMs infiltration was significantly lower than that of patients with low TAMs infiltration,and the difference was statistically significant(P<0.001).There was no significant correlation between metastasis and TNM stage(all P>0.05);the 5-year survival rate of patients with high TANs infiltration was significantly higher than that of patients with low TANs infiltration,and the difference was statistically significant(P<0.001).Conclusion As important immune cells in the TME,TAMs and TANs are involved in the occurrence and development of gastric cancer and may become potential targets for gastric cancer therapy.
