Objective To investigate the effects of exogenous hydrogen sulfide on body weight and lipid metabolism in obesity mice.Methods Male C57BL/6J mice were selected to establish DIO mouse model.After successful model construction,mice fed with low-fat diet and high-fat diet were randomly divided into four groups according to different intervention methods.They were low-fat diet control group(group A),diet-induced obesity(DIO)model control group(group B),50umol/(kg·d)Na HS treatment intervention group(group C1)and 100umol/(kg·d)Na HS treatment intervention group(group C2).Continuous intervention(intervention for 2,4,6 and 8 weeks respectively as planned)was conducted.During the intervention,the type of feed remained unchanged.The body weight of the mice was measured and recorded every 2 weeks.The body weight,epididymal fat weight and lipid metabolism of mice in each group were analyzed.Results Compared with the low-fat diet group,DIO model was established successfully,and body weight,epididymal fat weight,total cholesterol and triglyceride were significantly increased,while liver weight did not change significantly.Compared with DIO model control group,100umol/(kg·d)Na HS could aggravate liver steatosis in obese mice and increase fat accumulation in epididymal adipose tissue cells.Na HS treatment with 50umol/L(kg·d)for 8weeks can significantly improve hepatic steatosis in mice fed with high fat diet,but has no significant effect on fat accumulation in epididymal adipose tissue.The serum triglyceride levels were decreased after 4 weeks of Na HS intervention with 50 and 100umol/(kg·d).Conclusion(1)In this study,it was found that exogenous H2S intervention had no significant effect on the body weight of DIO mice.(2)This study found that intervention with exogenous hydrogen sulfide could promote the remodeling of epididymal adipose tissue in DIO mice,and this remodeling may be related to the beige of white fat.(3)This study proved that intervention with exogenous hydrogen sulfide can influence the ectopic fat accumulation in liver of DIO mice.
