Expression Of Long-stranded Non-coding RNA LINC00662 In Hepatocellular Carcinoma And Its Clinical Significance

Objective:To investigate the expression level of long non-coding RNA（lncRNA）LINC00662 in hepatocellular carcinoma（HCC）and its clinical significance,to investigate the effect of LINC00662 on the proliferation of HCC cells,and to explore the role of LINC00662 in the occurrence and development of HCC.Methods:（1）The R software was used to analyze the RNA sequencing data and follow-up information of patients with hepatocellular carcinoma from The Cancer Genome Atlas（TCGA）database,to compare the differences in LINC00662 expression between hepatocellular carcinoma tissues and normal tissues adjacent to the carcinoma,and to draw survival analysis curves using the Kaplan-Meier method to analyze the relationship between LINC00662 expression level and prognosis of hepatocellular carcinoma patients was analyzed by Kaplan-Meier method.（2）Seventy patients with hepatocellular carcinoma who underwent radical surgical resection from February 1,2021 to July 1,2021 in the First Bethune Hospital of Jilin University were selected,and the expression levels of LINC00662 in cancer and paraneoplastic tissues of these patients were measured by reverse transcription PCR（RTPCR）to compare the expression differences.Pearson χ2 test was used to investigate the relationship between LINC00662 expression and the clinicopathological characteristics of the patients.（3）A plasmid（sh RNA）was designed to interfere with LINC00662 expression and transfected with SMMC-7721 and HepG2 cell lines using transient transfection technique.The transfection efficiency was observed using fluorescence microscopy and the knockdown results were verified using RT-PCR,while a blank control group（sh-NC group）has been set up.（4）Analysis of the effect of LINC00662 on the proliferation of hepatocellular carcinoma cells using the Cell Counting Kit-8（CCK-8）.Result:（1）Based on the analysed data from the TCGA database,LINC00662 expression was significantly upregulated in hepatocellular carcinoma tissues（P<0.001）and patients with high LINC00662 expression showed a relatively poor prognosis（P<0.01）.（2）The up-regulated expression of LINC00662 in hepatocellular carcinoma tissues was verified by clinical tissue samples,and the expression level of LINC00662 was positively correlated with the degree of tumor differentiation,vascular invasion and lymph node metastasis.（3）LINC00662 expression was significantly higher in hepatoma cell lines（SMMC-7721 and HepG2）than in normal liver cell lines（Lo2）.（4）Down-regulation of LINC00662 expression in SMMC-7721 and HepG2 cell lines significantly inhibited the proliferation of HCC cells.。Conclusion:（1）LINC00662 is highly expressed in hepatocellular carcinoma tissues and cell lines,and the high expression of LINC00662 is associated with poor prognosis of HCC patients.（2）The expression level of LINC00662 in HCC was positively correlated with tumor differentiation,vascular invasion and lymph node metastasis.（3）Down-regulation of LINC00662 expression can inhibit the proliferation of HCC cells.
