ObjectiveTo investigate the expression and clinical significance of CGN in endometrioid carcinoma and to study the association between CGN expression and clinicopathological characteristics and prognosis of endometrioid carcinoma.Also aim to explore the possible mechanism of CGN,in order to provide new ideas and directions for the early diagnosis and treatment of endometrioid carcinoma.MethodPreliminary study on the differential expression of CGN in normal endometrial during hyperplasia tissues and endometrioid carcinoma tissues in TCGA、GEPIA and Oncomine databases.Meanwhile select patients in normal endometrial during hyperplasia,endometrial hyperplasia and endometrioid carcinoma who was confirmed by pathological examination in the Central Hospital Affiliated to Shenyang Medical College and the Heping Branch of the PLA Northern Theater Command General Hospital from January 2018 to January 2021,and from each type select 30 cases with complete pathological and clinical data.The immunohistochemical method was used to detect the expression of CGN in different endometrial tissues,and the Fisher accurate probability method was used to analyze the clinicopathological parameters of endometrioid carcinoma(age,degree of differentiation,tumor size,myometrium invasion,lymph node metastasis,FIGO clinical stage).Collated and analyzed the survival information of patients in TCGA and GEPIA database,plot the survival curve,to verify the relationship between CGN expression and prognosis of patients.The related genes co-expressed with CGN were searched by Linked Omics database for analysis.The protein interaction model of CGN was constructed by STRING database.The relationship between CGN and endometrioid carcinoma immune cell infiltration was explored using TIMER database.UALCAN database was used to investigate the methylation of CGN in endometrial carcinoma.Result1.According to GEPIA database,CGN is abnormally expressed in a variety of tumors.2.According to TCGA database,CGN expression was up-regulated in endometrial cancer tissues compared with normal endometrial tissues,suggesting that cingulate protein may promote the development of endometrial cancer.3.The positive expressions rates of CGN in normal endometrial during hyperplasia were 10%(3/30),the positive expressions rates of CGN in endometrial hyperplasia were 20%(6/30),the positive expressions rates of cingulin endometrioid carcinoma were 70%(21/30).Respectively,with statistically significant differences among the three groups(P < 0.05).4.In clinicopathological features,the expression CGN was closely correlated with muscular infiltration and lymph node metastasis of endometrioid carcinoma patients(P < 0.05).5.According to TCGA and GEPIA databases,the analysis of CGN and the survival and prognosis of patients with endometrial cancer showed that high expression level of CGN was significantly correlated with the poor prognosis of patients,suggesting that CGN may be related to the long-term survival of patients with endometrial cancer.6.The co-expressed genes of CGN were analyzed by using the Linked Omics database,and the results showed that CGN and these co-expressed genes participate in biological processes such as biological regulation,cell proliferation and growth,cell metabolism and stimulation responses through protein,ion,nucleic acid binding and various enzymatic reactions.It is closely related to insulin signaling pathway,inositol phosphate metabolism pathway and TNF signaling pathway.7.The CGN interaction model constructed by STRING database suggested that the 10 proteins most closely interacting with CGN were mainly involved in the formation and regulation of paracellular permeability barrier,cell cycle regulation,antigen presentation,immune cell activation,neutrophil transport and other biological functions.8.TIMER database analysis results showed that CGN had a significant positive correlation with neutrophil(r=0.228,P <0.01),which was statistically significant.9.By using the UALCAN database,which is found that CGN methylation is irrelevant to endometrioid carcinoma.10.CGN may be a oncogenic factor and a potential prognostic biomarker of endometrial adenocarcinoma.These studies provide new ideas for the pathogenesis of oral light protein and anti-tumor targeted therapy.ConclusionThe expressiongs of CGN in endometrioid carcinoma tissues were significantly higher than those in normal endometrial during hyperplasia and endometrial hyperplasia.CGN was closely correlated with the degree of muscular infiltration and lymph node metastasis of endometrioid carcinoma.To sum up,CGN may be a cancerpromoting factor and potential prognostic biomarker of endometrioid carcinoma.These studies provide a theoretical basis for pathogenesis of endometrioid carcinoma and antitumor targeted therapy.
