ObjectiveFrom a global perspective,colorectal cancer(CRC)currently ranks third in the incidence of malignant tumors,and its mortality ranks fourth in malignant tumors.Although the level of clinical CRC diagnosis and treatment has improved with the continuous improvement of the medical level of human society in recent years,the 5-year survival rate of CRC patients has not improved significantly.Therefore,if we want to achieve early detection and early diagnosis of CRC,early treatment and good prognosis and other goals,still a long way to go.Ubiquitin-proteasome system(UPS)widely exists in various cells of human tissues,and its essential function is to degrade intracellular cyclins by consuming ATP energy,so as to maintain the intracellular environment of the body.effective stability.Ubiquitin-conjugating enzymes E2C(UBE2C)plays an extremely important role in the UPS.Its main role is to regulate the mitotic step in the process of cell growth in eukaryotes,thereby affecting the cellular Growth cycle takes effect.According to relevant research and literature,once UBE2 C is highly expressed in cells,the degradation process of related cyclins through UPS will be disturbed,which will increase the instability of chromosomes,resulting in a large number of chromosome segregation errors.In this way,there will be isolated cells with abnormal chromosomal content,and the final result is often to promote the occurrence of malignant tumors.Some studies have clearly pointed out that UBE2 C has been highly expressed in many malignant tumors.However,as far as the current situation is concerned,there are very few studies at home and abroad on the correlation between the expression of UBE2 C and colorectal cancer.This study will detect the expression of UBE2 C in CRC tissue cells and adjacent tissue cells by immunohistochemical staining of specimen tissue sections,and then systematically analyze its expression in colorectal cancer tissue.To further determine whether it has a certain correlation with the prognosis of patients after CRC surgery.This will enrich the research status of the relationship between UBE2 C and colorectal cancer at home and abroad,and is likely to provide new treatment ideas for the future clinical treatment of CRC.MethodsThe surgical resection specimens of CRC patients who underwent surgical treatment in the General Surgery Department of the Affiliated Central Hospital of Shenyang Medical College from January 2011 to January 2021 were systematically collected.For example,paraffin tissue blocks are made.Ensure that all patients have not received neoadjuvant radiotherapy and chemotherapy before surgery,and have signed the relevant informed consent.Before the experiment,the paraffin tissue blocks were made into paraffin tissue sections,and immunohistochemistry(IHC)was performed on the tissue sections to detect the expression of UBE2 C in CRC tissue cells and adjacent tissue cells,and analyze UBE2 C according to the experimental results.Whether there is a correlation between the expression of and the clinical case data indicators and prognosis of colorectal cancer patients.ResultsAccording to the experimental results,UBE2 C showed high-intensity positive expression in the nucleus of CRC tissue cells,and slightly positive expression in the intercellular substance and cytoplasm;The expression rate in cancerous tissue cells was significantly higher than that in adjacent cancerous tissues(P<0.05);There was a positive correlation between UBE2 C expression and histological grade of cancer cells,depth of invasion,degree of lymph node metastasis,and TNM stage(P<0.05);There was no obvious correlation between UBE2 C expression and sex,age,tumor site,and tumor type of CRC patients(P>0.05);The results of Kaplan-Meier survival curve analysis indicated that the postoperative survival time of CRC patients with high expression of UBE2 C was significantly shorter than that of low expression patients(P<0.05);Cox univariate and multivariate regression analysis showed that UBE2 C expression acts as an independent postoperative prognostic factor for CRC patients(P<0.05).Conclusions1.UBE2 C is highly expressed in colorectal cancer tissues.2.There was a positive correlation between UBE2 C expression and histological grade of cancer cells,depth of invasion of cancer foci,degree of lymph node metastasis,and TNM stage.3.UBE2 C is an independent factor that can be identified as an independent factor for assessing postoperative prognostic status in patients with colorectal cancer.4.UBE2 C will be a potential target for CRC treatment.
