| Object:The periodontitis model of SD rats was established by the method of "ligation +high sugar diet +(P.g.)inoculation + prednisolone acetate injection".After the intervention of Jiawei Buzhong Yiqi Decoction,Gingival index(GI),plaque index(PLI),periodontal pocket depth(PD),gingival crevicular bleeding index(SBI),matrix metalloproteinases(MMP-1,MMP-2,TIMP-1,TIMP-2),bone biological parameters(Tb.N,Tb.SP,BV/TV)and other indexes were detected.To investigate the effect of supplementing qi and Invigorating Yang and strengthening teeth on bone destruction in SD rats with experimental periodontitis.Methods:1.A periodontitis rat model was established and a preliminary experiment was conducted: 20 SPF SD rats were selected at the age of 8 weeks,male and female,body weight 220-250 g,respectively.According to body weight,8 rats were randomly divided into blank group(Control-P group),Model group(Model-P group),and prevention group(BZYQ-P group).After 2 weeks of adaptive feeding,the Model-P group and BZYQ-P group were established by the method of tooth neck silk ligation+ high glucose +(P.G.)bacteria inoculation + prednisolone acetate injection to establish the animal Model of chronic periodontitis.From the 4th week,BZYQ-P group was given the preparation solution of Jiawei Buzhong Yiqi Decoction 10.5g/kg by gavage,once a day,for 4 weeks continuously,and the model was built continuously during the administration.The other feeding conditions were the same.Four rats in the BZYQ-P group were killed at 8 weeks after modeling,and four rats in the Control-P group and Model-P group were killed at 4 weeks and 8 weeks after modeling,respectively.The indexes related to periodontitis,HE staining and 3D reconstruction of alveolar bone were detected.2.Study on the prevention of experimental periodontitis in SD rats with the method of supplementing qi and Invigorating Yang and strengthening teeth: 4 rats were randomly selected in the prevention group(BZYQ-P),with half male and half female;From the 4th week,the BZYQ-P group was given Jiawei Buzhong Yiqi Granules 10.5g/kg by gavage,once a day,for 4 consecutive weeks,during which the model was built.The other feeding conditions were the same.Four rats in the BZYQ-P group were killed after 8 weeks of model building.The indexes related to periodontitis and HE staining were detected.3.Study on the effect and mechanism of supplementing qi and Invigorating Yang and strengthening teeth destruction of experimental periodontitis in SD rats: A total of 88 SD rats,32 rats without model and 56 rats with model were selected.The rats were numbered according to their body weight and randomly divided into 3groups in equal parts male and female.There were 32 patients in Control group(Control-T),32 patients in Model group(Model-T),and 24 patients in BZYQ-T.From the 8th week,the BZYQ-T group was intragastrically administrated with the preparation solution of Jiawei Buzhong Yiqi Decoction at a concentration of 10.5g/kg,once a day,for continuous 4 weeks.The Control-T group and Model-T group were intragastrically administrated with the same amount of normal saline,and the other feeding conditions were the same.Eight rats in the control-T group and the Model-T group were sacrificed at 0,1,2 and 4 weeks,respectively.Eight rats in the Bzyq-T group were sacrificed at 1,2 and 4 weeks,respectively.Periodontal tissues of the right maxilla were separated,and pathological specimens were prepared.Immunohistochemical staining was performed on the paraffin sections of the periodontal pathological specimens by matrix metalloproteinases MMP-1,MMP-2,TIMP-1 and TIMP-2,and image quantitative analysis was performed.The periodontitis-related indicators(GI,SBI,PLI,PD)were detected.Micro-CT was used to analyze the bone volume fraction,trabecular number and trabecular separation of the first molar of the right maxilla.Results:1.The periodontitis Model of SD rats was established by the method of "ligation+ high sugar diet +P.g.inoculation + prednisolone acetate injection".Compared with the Control group,the expression of GI,SBI,PLI and PD in the Model group improved after 4W.Compared with the Control group,GI,SBI,PLI and PD in the Model group were all higher after 4W,with statistical significance(P<0.05).Model group showed slight or mild necrosis of fibroblasts in the lamina propria of mucosa,accompanied by uneven neutrophils infiltration.Three-dimensional reconstruction of alveolar bone after 4W in the Model group showed obvious alveolar bone absorption,vertical absorption,exposed root,and reduced periodontal tissue attachment.2.Effects of Jiawei Buzhong Yiqi Decoction on MMP-1,MMP-2,TIMP-1and TIMP-2 in Periodontal Tissue:Compared with the Control group,the expression of MMP-1,MMP-2,TIMP-1and TIMP-2 were higher in the Model group at 0W,1W,2W and 4W,and the difference was statistically significant(P<0.05).Compared with the Model group,the expressions of MMP-1,MMP-2,TIMP-1and TIMP-2 were decreased at 4W of BZYQ,with statistical significance(P<0.05).3.Micro-CT analysis of the effects of Jiawei Buzhong Yiqi Decoction on periodontal related indexes and alveolar bone resorption in SD rats:Compared with the Control group,the expression of GI,SBI,PLI and PD were higher in the Model group after 0W,1W,2W and 4W,with statistical significance(P<0.05).Compared with the Model group,the expressions of GI,SBI,PLI and PD in the BZYQ group were all decreased after 4W,with statistical significance(P<0.05).Compared with the Control group,the expression of BV/TV and Tb.N were lower in the Model group at 0W,1W,2W and 4W,while the expression of Tb.SP was higher,with statistical significance(P<0.05).Compared with Model group,the expression of BV/TV in BZYQ group was increased and the expression of Tb.SP was decreased at 4W,with statistical significance(P<0.05).Conclusion:1.The method of tonifying qi and raising Yang to strengthen teeth can reduce the inflammatory response of periodontal tissue in SD rats with experimental periodontitis,prevent the destruction of alveolar bone in rats with periodontitis,and reduce alveolar bone absorption. |
