Objective:Based on the“jin“ for bone of physiological and pathological basis,by observing the“muscle“ technique for knee joint model of rabbit cartilage pathological changes and cytokine TNF alpha,MMP-3,TIMP-1 and TGF-beta 1 in different groups of rabbit articular fluid exists in the content is the difference between how much,attempt from the perspective of cytokines shallow exploration “muscle“ technique in the treatment of knee osteoarthritis at the time of the working mechanism,in order to clinical front line staff to provide more treatments as well as the corresponding theoretical basis.Research methods:The random number table method was used to select 12 healthy rabbits as blank group,and no modeling treatment was given.Then 38 rabbits were selected to model the right hind leg,and 4% papain solution was injected through the articular cavity in different times according to the modeling requirements.After the modeling,2 rabbits were randomly selected for modeling verification.After the modeling was successful,the other rabbits were randomly divided into 3 groups with 12 rabbits in each group,which were successively named model group,control group(referred to as control group)and “jin relieving“ manipulation group(referred to as manipulation group).At the end of grouping,rabbits in each group were given corresponding intervention,as follows: blank group and model group were not given intervention;The control group was injected with sodium hyaluronate once a week.Manipulation group was treated with “relieving tendons“ every other day.Four groups were investigated respectively according to their group intervention intervention plans,and then at the end of the second month of the intervention and the fourth month 6 random executed the rabbit,respectively,and materials,processing,and overall state respectively to observe the rabbit articular cartilage,knee joint state and general form and complete the scale,then using enzyme-linked immunoassay determination of TNF alpha in joint fluid,MMP-3,TIMP-1 and TGF-beta 1 content,records,analyze data,draw the conclusion.Results:1.The knee joint scale evaluation of the modified Lequesnemg and the morphological scores of articular cartilage under gross observation were as follows: compared with the blank group,the scores of the model group,the control group and the manipulation group at the end of the second and fourth months all increased(P < 0.05);However,compared with the model group,the scores of control group and manipulation group were decreased(P < 0.05).There was no significant difference between the two treatment groups(P > 0.05),but the content of each group at the end of the fourth month was still decreased compared with that at the end of the second month(P < 0.05).2.Enzyme-linked immunoassay determination of TNF alpha in joint fluid,MMP-3,TIMP-1 and TGF-beta 1 content: compared with the blank group,model group,control group and the content of the cytokine levels were increased(P < 0.05),but compared with model group,the other two groups after give corresponding treatment intervention on rabbit,cytokine levels have decreased(P < 0.05).There was no significant difference between the two treatment groups(P > 0.05),but the content at the end of the fourth month in each group decreased compared with that at the end of the second month(P < 0.05),indicating that the treatment effect of the two treatment groups improved with the extension of treatment time,but there was no significant difference in the efficacy between the two groups.Conclusion:1.The "Shujin" technique could significantly reduce the evaluation of knee joint scale Lequesne MG modified KOA model of rabbits and the morphological score of articular cartilage observed with naked eye,as well as the TNF-α、MMP-3、TIMP-1 and TGF-β1 content in articular fluid,and adjust the MMP-3/TIMP-1 ratio to the normal rabbit level in blank group.2.All the indexes of Shujin manipulation group and control group were similar,and the effect of Shujin manipulation and sodium hyaluronate joint cavity injection in early KOA was similar. |
