| Objective: Although long non-coding RNA(lnc RNA)LINC01134 has been identified as a crucial regulator in hepatocellular carcinoma(HCC),and no relevant study has reported the correlation between LINC01134 and breast cancer(BC).In the present study,we aimed to explore its expression and biological functions in BC.Methods: Relative LINC01134 expression in 96 pairs of BC tissues and adjacent normal tissues were detected using q PCR analysis.SPSS software was used to analyze the correlation between the expression level of LINC01134 and clinicopathological parameters such as tumor size,TNM stage and lymph node metastasis.The role of LINC01134 in BC cell biological functions were explored using CCK-8 cell proliferation assays,colony formation assays,transwell migration and invasion assays,respectively.The molecular mechanisms through which LINC01134 regulated BC cell proliferation and metastasis were confirmed by q PCR and western blot analyses.Results: We demonstrated that LINC01134 expression was significantly higher in BC tissues than in adjacent normal tissues,the expression of LINC01134 in breast cancer was significantly higher than that in adjacent tissues.The up-regulated expression of LINC01134 in breast tumors is significantly correlated with the clinicopathological parameters such as tumor size,TNM stage,lymph node metastasis,etc.The higher expression level of LINC01134 in breast tumors indicates the shorter survival time and the worse prognosis of breast cancer patients.Data analysis showed that the expression of LINC01134 can be used as a predictor of prognosis in breast cancer patients.Knockdown of LINC01134 suppressed BC cell proliferation,migration and invasion in vitro.Molecular mechanism studies confirmed that LINC01134 plays a biological role in breast cancer by up-regulating the expression of β-catenin and its downstream related molecules.Conclusion: In this study,we identified a novel oncogene LINC01134 that promotes the proliferation,migration and invasion of breast cancer through activatin theβ-catenin.The expression level of this gene was significantly associated with the prognosis of breast cancer patients.This study shows that LINC01134 can be used as a potential prognostic biomarker and therapeutic target for breast cancer. |
