| Objective:To explore the expression of Ig G4 and STAT5 b in normal middle turbinate and chronic rhinosinusitis with nasal polyps and their correlation with clinicopathological features.Immunofluorescence localization and Western blotting(Western Blotting,WB)were used to quantify the expression in nasal polyps.At the same time,the recruitment of immune cells and the changes of inflammatory factors in the pathological microenvironment of chronic rhinosinusitis with nasal polyps were discussed.Methods:Paraffin sections were used for routine pathological staining and Sirius red staining,and frozen sections were used to localize the expression of Ig G4 and STAT5 b in normal middle turbinate and chronic rhinosinusitis with nasal polyps.The protein extracted from normal middle turbinate and nasal polyps was tested by WB to quantify the expression of Ig G4 and STAT5 b in normal middle turbinate and nasal polyps.At the same time,neutrophils and macrophages in the pathological microenvironment of nasal polyps were visualized by immunofluorescence,and the m RNA levels of inflammatory cytokines TNF-α,IL-1 β,IL-6 and chemokine CCL4,CCL5,CCL14 in the two tissues were detected by PCR.Results:Compared with the normal middle turbinate tissue,the glands in the pathological tissue of nasal polyps were reduced and showed a state of fibrosis,and Ig G4 and STAT5 b were highly expressed in nasal polyps.In addition,neutrophils and macrophages were recruited in the pathological microenvironment of nasal polyps,and the m RNA levels of inflammatory factors TNF-α、IL-1 β、IL-6 and chemokines CCL4、CCL5 and CCL14 were significantly increased.Conclusions:The expression of Ig G4 and STAT5 b is increased in nasal polyps,and the pathology of nasal polyps is in a state of fibrosis.the occurrence and development of nasal polyps is closely related to the pathological characteristics of fibrosis.The recruitment of neutrophils and macrophages and the participation of a large number of inflammatory factors and chemokines in nasal polyps indicate that nasal polyps show a complex pathological microenvironment.The inflammatory microenvironment of nasal polyps may play an indicative role in the pathological definition of nasal polyps.At the same time,Ig G4 and STAT5 b are expected to provide a new index for clinical screening,diagnosis,treatment and prognosis of nasal polyps,as well as a new theoretical basis for later targeted treatment of nasal polyps. |
