Objective:By observing the cell morphology and proliferation activity of RPE cells after blue light irradiation,and identifying and analyzing the differentially expressed genes of RPE cells after blue light irradiation,we can deeply understand the mechanism of AMD induced by blue light,and explore the potential targets for the treatment of AMD.It provides a new idea and science for repairing RPE injury induced by blue light.Methods:(1)ARPE-19 cells were divided into two groups: control group(dark)and blue light group(2,4,6,8 h).The cell morphology was observed under microscope,and the proliferation rate of ARPE-19 cells was detected by CCK-8 method.(2)Second generation sequencing was performed in the blue light group exposed to 0,4 and 6h light irradiation,and the m RNA levels of each group were detected by q PCR.Results:(1)The effect of blue light on the morphology of ARPE-19 cells: the morphology of ARPE-19 cells changed after 4 hours of blue light irradiation,and the cells became smaller and rounder;the intercellular space became larger after 6 hours of blue light irradiation,and the number of abnormal cells increased;cell debris could be seen in the culture solution after 8 hours of blue light irradiation,and the number of abnormal cells increased further compared with 6 hours.(2)Effect of blue light on the proliferation rate of ARPE-19 cells: the experiment was divided into control group(dark group)and blue light groups with different light duration(2h,4h,6h,8h);CCK-8 method was used to detect the proliferation rate of ARPE-19 cells in each group.Compared with the control group,the proliferation rat e of each blue light group was decreased(P < 0.01),and the proliferation rate was negatively correlated with the light duration(P < 0.01).(3)The transcriptome was sequenced at 0,4 and6 h,The results were confirmed by q PCR.APOE gene was the most significantly up-regulated gene and TMEM129 gene was the most significantly down-regulated gene.Conclusion:(1)Blue light irradiation on ARPE-19 cells in vitro can cause damage and morphological changes.(2)Blue light irradiation can change the proliferati on activity of ARPE-19 cells in vitro,resulting in the decrease of proliferation activity.(3)Blue light irradiation can induce differential gene expression in ARPE-19 cells.
