Panax Notoginseng(PN)and Salvia Miltiorrhiza(SM)extracts have an extraoridinary antioxidant properties,which have been widely used in pharmaceutical industry,cosmetics industry and food preservation.However,the current methods to extract Chinese herbs still have some shortcomings,including low efficiency and low utilization rates.This paper mostly focuses on the method combined continuous extraction with several separation technologies,including membrane separation and column separation.The yield and the purity of active ingredients,antioxidant properties and other factors have been taken into consideration to choose the more appropriate sparation technology during the course of building up a comprehensive,efficient,environmentally-friendly and simply methods to extraction and separation.Besides,this paper preliminarily expored the relation between active ingredients and antioxidant properties.We can get the following conclusion:(1)Continuous extraction had taken effect in further extraction of active ingredients and full utilization of Chinese herds.Analysing through several chemical analysis methods,there are saponins,polysaccharide and amino acid in PN extracts,whereas there are phenols,polysaccharide and amino acid in SM extracts.Separating PN and SM extracts by D101 macroporous resin chromatographic column has higher yield and purity of PN saponins and SM phenols.Combining the concentration by 150 Da membrane and separation by alcohol sedimentation has the highest purity of PN and SM polysaccharide,which is 90.3% and 97.3%,respectively.(2)Considering from antioxidant properties,the intercepting part,separating by 3500 Da membrane has higher reducing ability,and therefore its antioxidant properties is better.Concentrating the second PN extract by 150 Da membrane is efficient and the intercepting part has high ability to scavenge hydroxyl radical and higher reducing ability.Moreover,the ability to scavenge DPPH radical of those products from PN aren’t evident.The product separating from the first SM extract by membrane method don’t have better antioxidant properties.The 30% ethanol eluting part,separating from the second SM extract,has better antioxidant properties.The intercepting part,concentrating from the third SM extract by 150 Da membrane,has slightly better antioxidant properties.(3)Considering from the relation between active ingredients and antioxidant properties,the purity of PN polysaccharide correlated with the reducing ability whereas the purity of SM phenols positively correlated with the reducing ability and the ability to scavenge DPPH radical.(4)Analysing through SH-SY5 Y cell experiment,the intercepting part,separating from the first PN extract by 3500 Da membrane and the intercepting part,separating from the second SM extract by 3500 Da membrane is beneficial to the survival rate of injured cell by hydrogen peroxide.(5)Analyzing through response surface method,the drug pair of PN and SM has mostly antagonistic effects but there is slight synergistic effect at low cocentrations.
