Objective: To investigate the effect of Alda-1 on cytokines of radiation pneumonia in rats.Methods:(1)Fifteen male SD rats were randomly divided into two groups: the control group(n=5)and the model group(n=10).After determining the irradiation field,the model group received 20 Gy one-time irradiation of the whole chest,while the control group received no irradiation.The clinical manifestations of the rats were recorded throughout the experiment,and lung CT examination was performed 8 weeks after irradiation to observe the inflammation and fibrosis.(2)Seventy-six SD rats were randomly divided into 5 groups:ALDA-1 treatment group(n=20),dexamethasone treatment group(n=12),Daidzin intervention group(n=20),model group(n=12),and normal control group(n=12).The normal control group received feint irradiation,and the remaining 4 groups were positioned under CT,and the whole chest was irradiated with 20 Gy once after the irradiation field was defined.After irradiation,medication was given once a day.The model group and the control group were respectively infused with the same amount of normal saline.Pulmonary CT examination was performed at the weekend 1 and4 after irradiation(1/2 rats were randomly selected from each group at the weekend 1 and 4),and 3 imaging diagnostics experts in our hospital were randomly selected to make imaging diagnosis by blind method and grade according to clinical manifestations.After imaging examination,the rats were sacrificed and blood samples were collected to determine the concentrations of TNF-α,TGF-β and IL-6 in serum.Results :(1)The general condition and lung CT of the rats in the control group were normal,while the rats in the model group showed reduced activity,dyspnea and depilation in the irradiation field,and lung CT showed different degrees of exudation and fibrosis.No death occurred during this process.(2)Serum concentrations of TNF-α,TGF-β and IL-6 in model group were higher than those in blank group(P<0.05);The concentrations of TNF-α,TGF-β and IL-6 in ALDA-1 group and dexamethasone group were significantly lower than those in model group(P<0.05);Serum concentrations of TNF-α,TGF-β and IL-6 showed no significant differences among the treatment groups(P>0.05);Serum concentrations of TNF-α,TGF-β and IL-6 in Daidzin group were significantly higher than those in model group(P<0.05).(3)The general condition and lung CT of normal control group rats were normal;After irradiation,the rats in the other groups had less movement,less food intake,less water intake,different degrees of depilation and dyspnea in the irradiation field,and the order of severity was:Daidzin group>Model group>Dexamethasone group >Alda-1.During the experiment,1 rat died in the Daidzin group and 1 rat died in the dexamethasone group.After irradiation,lung CT of each group showed different degrees of inflammation and fibrosis,and the severity of the changes was as follows:Daidzin group>Model group>ALDA-1 group =dexamethasone group.Conclusion :(1)Through CT simulation location,20 Gy one-time irradiation of the whole chest of rats can successfully establish a radiation pneumonia model;(2)ALDA-1 can delay the occurrence and development of radiation pneumonia,the mechanism of which may be through the enhancement of ALDH2 activity to eliminate ROS in vivo,reduce the expression of TNF-α,IL-6,TGF-β and other inflammatory factors,and reduce the inflammatory response. |
