Objective:To explore the effects of VEPH1(Ventricular zone-expressed PH domain-containing protein homolog 1)on the regulation mechanisms of skin melanoma(Cutaneous melanoma,CM)cell cycle,epithelial-mesenchymal transition(EMT)and apoptosis the study.Methods:A375,B16-BL6,B16 melanoma cell lines and human normal skin cells(HaCaT)were selected for cell culture in vitro,and the mRNA expression level of VEPH1 was detected by real-time fluorescence quantitative polymer chain reaction(q RT-PCR).The screening is of the most experimental significance.The melanoma cell lines were transfected and grouped into 8 groups: blank group(untreated),negative control group(transfected with blank plasmid),VEPH1 group(transfected with overexpressing VEPH1 plasmid),SB-431542 group(transfection of TGF-β receptor inhibitor),VEPH1+SB-431542 group(co-transfection of TGF-β signaling pathway inhibitor and overexpression of VEPH1plasmid),TGF-β group(transfection of overexpression of TGF-βplasmid),VEPH1+TGF-β group(co-transfected with over-expressing TGF-β plasmid and over-expressing VEPH1 plasmid),si-VEPH1 group(transfected with si-VEPH1 plasmid),using qRT-PCR,Western Blot to detect each group of transfected cell lines The expression levels of mRNA and protein of VEPH1,Smad4,TGF-β,EMT-related proteins in VEPH1,Smad4,EMT-related proteins;flow cytometry was used to detect the cell cycle and apoptosis of each group of transfected cell lines.Conclusion:VEPH1 is low expressed in skin melanoma;overexpression of VEPH1 can inhibit skin melanoma cell cycle and EMT through TGF-βsignaling pathway,and promote skin melanoma cell apoptosis.
