Objective: Selaginella,as one of the rare plants belonging to single family and single genus,has a history of 400 million years,so it is known as a living fossil in the process of plant evolutionary.Selaginella has been widely used as an herbal medicine in many areas nowadays,and it is included as a medicinal material in 2020 Pharmacopoeia.In recent years,Selaginella has attracted increasing attention of natural researchers because it contains a large number of new structural compounds.In order to obtain a single medicinal ingredient with novel structure and significant anti-hepatoma activity,this paper separate and purify the chemical components of Selaginella by modern chromatographic separation technology.Moreover,evaluate their anti-hepatocarcinoma activity through in vitro activity screening,and provide for the elucidation of the anti-tumor medicinal substances of Selaginella Experimental basis.Methods: Based on the preliminary work of our research team and the judgment of ultraviolet characteristic spectrum,this paper selects the n-butanol part of Selaginella for the research object of this subject.The n-butanol part of Selaginella is separated and purified by modern extraction and separation techniques,such as extraction,column chromatography and HPLC.Dentify monomer compounds by UV,nuclear magnetic resonance spectroscopy and high-resolution mass spectrometry.The CCK-8 experiment was used to determine the effect of monomeric compounds on the viability of liver cancer cell lines,so as to screen for compounds with anti-hepatoma effect.Results: In this paper,22 compounds,including 12 new compunds,were isolated and identified from the n-butanol part of Selaginella.Among them,compound 1* is a rare ring-opening biphenyl anthrone derivative in nature,compound 2* is isobenzene.Parafuran derivatives,compounds 3*,4*,5*,6*,7*,8* are new acetylenic phenol derivatives,compound 9* is a diaryl substituted stilbene ketone derivative,compound10*,11*,12* are benzophenone derivatives,and compound 10* is a rare diaryl benzophenone compound.In addition,we also obtained 10 known compounds,and a total of 22 monomer compounds were isolated from Selaginella.Molecular biology methods is used to screen the viability of selected compounds.Compounds 5*,6*,8*,16,22 have a certain inhibitory effect on the proliferation of hepatoma cells.Conclusion: In this paper,22 compounds have been isolated and identified,including 12 new compounds.Five compounds were screened to have the ability to inhibit the proliferation of hepatoma cell lines in vitro.
