| Background The number of people with asthma in the world is about 300 million,with the highest incidence reaching 18%.By 2025,the number of asthma patients worldwide is expected to increase by 100 million.The data of Global Burden of Disease Study showed that the incidence of asthma in China was 0.5-1.5% in 2015 with a total of 30 million people,which lead to more than 25000 people’s death and 1.8 million years of Disability Adjusted Life Year(DALY),ranking second,third and second in the world respectively.Allergic Asthma is common in infants and young people.It is most likely to be caused by Ig E antibodies,which is produced by allergens stimulating the body,Th cells also play an important role in this pathogenesis,expression level of T-bet and GATA-3 can also regulate the balance of Th1 and Th2 cells.As we all know,Astragalus have been widely used and studied for its anti-asthma action,however its mechanism of anti-asthma action is lack of deeply studied.Objective The aim of this study is to confirm the pharmacological effects of Astragalus on allergic asthma by establishing rat model of allergic asthma,on this basis the the effect of Astragalus on the expression of T-bet/GATA-3 in Lung Group is explored.Further more,we can ensure the mechanism of anti-asthma action in Astragalus’ and provide clinical research evidence for the prevention and treatment of asthma.Methods 60 eligible female rats were established by stimulating OVA twice(d1,d8)for 28 days,after that,corresponding drugs were given to each group of rats one hour before the stimulation for 28 days.30 allergic asthma model of SD rats with obvious symptoms were randomly divided into three groups: Group A,10 rats were inhaled fluticasone propionate;Group B,10 rats were taken Astragalus injection.Group C,10 rats were taken 0.9%NS as a control group.On the 28 th day of the experiment,bronchial provocation test(BPT)was performed.After recording relevant data,we executed animals within 24 hours after the last stimulation,and we can explore the possible anti-asthma action mechanism of Astragalus by observe pathological results T-bet and protein content detection of GATA-3.Results After successful modelling,there were no significant differences in weight,daily food intake,daily water intake,hair color and activity among each groups.At the end of experiment,rats in drug groups(Group A and Group B)gained more weight than those in group C(37.39±7.78 and 138.94±6.48 vs127.31±4.61,P=0.000).During sensitization stage,the allergic symptoms which are restlessness,shortness of breath,sneezing,nose scratching,trunk scratching and standing instability significantly most decreased compared with the control group.However,the difference between two drug groups(Group A and Group B)were not significant.Moreover the inflammatory cells in the pulmonary alveolus of drug groups(Group A and Group B)were less than that of control group,and the degree of bronchial mucosal edema and the deformations of tracheal as well as epithelial proliferation were also improved deeply.Western blots showed that there was no difference in the expression of T-bet protein between group A and group B(P > 0.05),but if that compared with Group C,significantly most increased(P < 0.05);The expression lever of GATA-3 protein in group A and group B had no differences(P > 0.05),but significantly decreased than that of group C(P < 0.05).Conclusion Astragalus injection can affect the balance between T-bet and GATA-3 by up-regulating the expression level of T-bet and down-regulating the expression level of GATA-3.It also play an important role in the treatment of asthma by up-regulating the differentiation of Th1 cells and down-regulating the differentiation of Th2 cells. |
