Objective: Osteosarcoma is one of the common bone-derived malignancies,accounting for about half of all bone cancers in the world,and is the second most fatal cancer among children and young people.Due to its rapid progress and prone to hemolymphatic metastasis This leads to a poor prognosis.Although in recent years great progress has been made in the treatment of osteosarcoma by surgical removal and multiple drugs combined with radiotherapy and chemotherapy,the 5-year overall survival rate of patients with osteosarcoma has increased to about 60%,but the overall survival of patients with late-diagnosed or with metastases tumors is still not ideal.Therefore,exploring the molecular mechanism that regulates the malignant progression of osteosarcoma and discovering effective molecules are of great significance for new treatments of osteosarcoma.This study aims to reveal whether the sirtuin SIRT5 plays an important role in regulating the malignant progression of osteosarcoma,that is,whether it regulates the proliferation,migration and invasion of osteosarcoma cells.Methods: First,collect normal bone tissues and osteosarcoma tissues of different stages from clinical related departments,and culture human normal osteoblasts h FOB1.19,osteosarcoma cell lines U2 OS and MG63.The expression levels of SIRT5 m RNA and protein in normal bone tissue,osteosarcoma tissue and osteosarcoma cell lines were detected by immunohistochemistry,quantitative Real-time PCR and Western blot methods.In order to further test its effect on the malignant biological behavior of osteosarcoma cells,a SIRT5 knockdown osteosarcoma cell line was constructed by using the stable transfection method of si RNA-SIRT5 to interfere with the expression of SIRT5,and the cell viability CCK-8 method and cell Transwell were used.Methods The proliferation,migration and invasion ability of osteosarcoma cells were detected respectively.Results: This study found that SIRT5 was significantly highly expressed in osteosarcoma tissues and cells.Knockdown of SIRT5 expression inhibits the proliferation,migration and invasion of osteosarcoma cells.Conclusions:1.SIRT5 is highly expressed in osteosarcoma tissues and is mainly distributed in the cytoplasm of osteosarcoma cells.2.SIRT5 is highly expressed in the osteosarcoma cell lines U2 OS and MG63.3.Knock down the expression of SIRT5 to inhibit the proliferation of osteosarcoma cells.4.Knock down the expression of SIRT5 to inhibit the migration and invasion of osteosarcoma cells.5.SIRT5 is correlated with the size of osteosarcoma in clinicopathological factors...
