| Objective To explore the expression level of CENPU in hepatocellular carcinoma(HCC)data set and the association between CENPU and prognosis of patients using bioinformatics methods,and to further study the expression level of CENPU in human HCC cell line in vitro.The effect of down-regulation of CENPU expression on the migration ability of HCCLM3 cells was studied.The relationship between CENPU and clinical prognosis of HCC patients was analyzed to explore the prognosis markers of liver cancer.Methods The m RNA expression of CENPU in Pan cancer was detected in UALCAN database,and then the protein expression of cancer and normal tissues was compared in CCLE.GEPIA and Kaplan-Meier Plotter were used to explore the relationship between the CENPU and clinical prognosis in HCC.The CENPU related interacting proteins was further gained from SRING database.Finally,the expression of CENPU was measured using q RT-PCR.Two lentivirus mediated sh RNA targeting CENPU were constructed,and then transfected into HCCLM3 cell line to obtain HCCLM3-sh CENPU-1# and HCCLM3-sh CENPU-2# as experimental groups,and NC group as control group wound healing assay was used to determine the migration ability before and after knockdown.The association between CENPU and clinical prognostic significance in HCC patients were evaluated by immunohistochemistry.Results Bioinformatics analyses showed that the expression of CENPU was significantly high in lots of malignant tumors The expression of CENPU in cancer tissues was significantly higher than that in adjacent cancer tissues.Through GEPIA,UALCAN,Kaplan Meier plotter and other databases to predict the expression of CENPU in HCC,we found that the expression of CENPU in HCC was significantly higher than in normal tissues,and the prognosis of patients with high expression of CENPU was poor.ROC analysis from GTEX online database found that CENPU has high prognostic diagnostic ability(AUC=0.926).The results from STRING showed that CENPU protein was closely related to CENPA,CENPO,CENPP,CENPK,CENPI,CENPQ,CENPR,CENPT and other proteins,and interacted with Plk1,CDK1,OIP5,ZWIN,BUB1,NDC80,ITGB3 BP and other proteins,and participated in the process of cell mitosis and cell cycle.Compared with cell line L02,the expression level of CENPU m RNA in HCCLM3,Hep G2 and Huh7 cell lines increased,and the highest expression level was in HCCLM3.The inhibitory effect of low expression of CENPU on cell migration(P<0.05)were found using HCCLM3-NC,HCCLM3-sh CENPU-1# and HCCLM3-sh CENPU-2#.The results of immunohistochemistry showed that the expression of CENPU was upregulated in HCC(P < 0.05).The expression of CENPU was not correlated with age,gender,pathological grade,tumor size and number,AFP expression,integrity of tumor capsule,infection of HBV and HCV,and biochemical indexes.Multivariate logistic regression showed that CENPU protein,pathological stage and GGT were independent prognostic factors for the overall survival of patients with HCC.Conclusion CENPU is highly expressed in HCC and is a tumor promoting gene.Low expression of CENPU inhibits the migration of HCCLM3 cells.The prognosis of HCC patients with high CENPU is poor. |
