Topiramate In Migraine Prevention: A Multicenter Observational Study In Southwest China

Objective: To assess the efficacy and safety of topiramate(TPM)as a migraine-preventive therapy in Southwest China.Methods: Patients were from the 7 headache centers in southwest China hospitals from April 2019 to June 2020.After a baseline period of 1month,and eligible patients were given topiramate treatment for 6 months.During the course,patients were required to keep detailed headache diary and regularly followed up every month to collect headache data.The primary endpoints was the 50% responder rate for monthly headache days after 6 months of treatment.The secondary endpointsincluded changes in monthly migraine days,Migraine Disability Assessment(MIDAS),Headche Impact Test-6(HIT-6)and adverse events.Results : A total of 50 patients were enrolled in the study,and 33 composed the intent-to-treat population.Migraine improved after topiramate-treated patients,but there was no significant in the 50mg/d group compared with baseline.Although the change of headache days was comparable in the 75mg/d group compared with the 100mg/d group,the50% responder rate in the 100mg/d group(59.09%)was higher than that in the 75mg/d group(42.11%)(P<0.05).After treatment,the most common adverse events was paresthesia(30%).Conclusion : Topiramate,75mg/d and 100mg/d,was effective as a preventive therapy for patients with migraine.
