Objective:(1)To explore the expression of mi R-181a-5p in gliomas.(2)To determine the effect of mi R-181a-5p targeting FBXO11 on the proliferation and invasion of drug-resistant gliomas.Methods:(1)In normal glioma cells(U251)and drug-resistant glioma cells(U251TR),the expression levels of mi R-181a-5p and FBXO11 m RNA were detected by RT-PCR;the expression levels of FBXO11 were detected by Western blot.(2)The mi R-181a-5p overexpression model was constructed in U251 TR cells and the mi R-181a-5p knockdown model was constructed in U251 cells and their corresponding NC and Control groups;CCK8 was used to detect cell proliferation;Cell scratch test and Transwell test was used to detect Cell migration and invasion ability;Cell apoptosis was detected by flow cytometry;FBXO11 expression was detected by RT-PCR and Western blot;finally,the dual luciferase reporter gene was used to verify the relationship between mi R-181a-5p and FBXO11 Targeted regulation association.Results:(1)Compared with U251 TR cells,the expression of mi R-181a-5p was reduced in U251 cells,and the expression of FBXO11 was increased.The difference was statistically significant(P <0.05).(2)Compared with the NC and Control groups,the U251 TR cells overexpressing mi R-181a-5p had decreased proliferation,migration and invasion capabilities,increased apoptosis,and decreased FBXO11 expression.The difference was statistically significant(P <0.05).(3)Compared with the NC and Control groups,U251 cells knocked down mi R-181a-5p had increased proliferation,migration and invasion abilities,decreased cell apoptosis,and increased FBXO11 expression.The difference was statistically significant Significance(P <0.05).(4)The results of dual luciferase reporter gene analysis show that mi R-181a-5p can target the3’untranslated region of FBXO11.Conclusion:(1)In drug-resistant glioma cells,mi R-181a-5p exhibits low expression.(2)As a tumor suppressor of glioblastoma,up-regulating mi R-181a-5p reduces cell proliferation and invasion,and promotes cell apoptosis.(3)When the expression level of mi R-181a-5p is up-regulated,it inhibits the protein expression of FBXO11 in drug-resistant glioma cells.
