Acne is a common chronic inflammation of hair follicles and sebum,which can have a negative impact on the patient’s mental health and social life.Treating acne with traditional Chinese medicine has the characteristics of pure nature,less toxic and side effects,good curative effect,and no dependence,and is favored by patients with skin diseases.Reasons such as clarity,inability to qualitatively and quantitatively,and inconvenience to carry have limited the development of traditional Chinese medicine.In order to solve this problem,two kinds of traditional Chinese medicine gels were prepared in this paper.The safety and effectiveness of the gel were determined by screening of traditional Chinese medicine,selection and optimization of gel,determination of physicochemical properties,determination of antioxidants,pharmacodynamics research and safety evaluation.At present,external medicines for the treatment of acne are mainly based on retinoids,benzoyl peroxide and antibiotics.The traditional Chinese medicine gel has many advantages such as good spreadability,easy cleaning,convenient use,and easy absorption through the skin.Its preparation process is relatively simple,and it is less restricted by medicinal materials.The main research contents and results of this paper are as follows:(1)Extraction of honeysuckle,dandelion,and violetine and preparation of gel.The traditional Chinese medicine was extracted by water extraction and alcohol precipitation,and two gels were prepared,namely carbomer gel and composite gel.(2)Optimization of gel matrix.Through a series of single factor experiments,the factors that have a greater impact on the matrix were found out and the response surface analysis was carried out,and the optimal material ratio of carbomer gel was obtained:1.39%of carbomer,10.18%of glycerol,3 Ethanolamine is 0.98%;the optimal material ratio of the composite gel is:0.13%of carbomer,0.1%of xanthan gum,and 0.4%of triethanolamine.(3)Determination of physicochemical indexes and evaluation of antioxidant activity of gels with different proportions of traditional Chinese medicines.The physical and chemical properties including pH,stability(heat resistance,cold resistance,centrifugation)and moisture retention were evaluated for the two gels,and the DPPH free radical scavenging rate was determined.The results showed that the pH values of each group of carbomer gel and composite gel were all around 6,which were in line with the national standard.The gel stability of each group was normal,and the sensory evaluation was good.In addition,the DPPH free radical scavenging rates of each group of carbomer gel were:54.0%,32.9%,47.2%,41.6%,57.4%,49.6%,52.8%;the DPPH free radical of each group of composite gel The clearance rates were:60.4%,39.2%,52.8%,46.2%,64.6%,56.8%,and 58.1%.Among them,the E group composite gel composed of honeysuckle,dandelion,and violacean(2:1:1)The best effect.(4)Pharmacodynamics study of gels with different ratios.According to the appearance of modeling,swelling degree,swelling inhibition rate and histopathological section observation,the therapeutic effects of three kinds of acne models established by gel p-xylene and croton oil were determined.The results showed that different proportions of gels had certain therapeutic effects on the three acne models.Judging from the swelling inhibition rate and the observation of pathological sections,the therapeutic effects were:E>A>G>F>C>D>B,the E group composite gel composed of honeysuckle,dandelion,and violetine(2:1:1)has the most obvious effect.(5)The research and safety evaluation of the antibacterial test of the two gels.The results showed that both gels had antibacterial activity,and the highest proportion of traditional Chinese medicines was in group E(Honeysuckle,Dandelion,Viola 2:1:1),and the composite gel was Bicarbomer Gel.The agent has good bacteriostatic ability.The safety evaluation showed that the two gels were non-irritating,non-sensitizing,safe and effective.In this experiment,two kinds of gels,carbomer gel and composite gel,were prepared,and their optimal gel ratios were screened out.Carbomer gel.Through a series of physical and chemical property determinations,pharmacodynamic studies and safety evaluations,two safe and effective gels were obtained that met the requirements of the Chinese Pharmacopoeia.
