| Objective Dendritic cells(DCs)are mainly derived from CD34+pluripotent hematopoietic stem cells(CD34+HPC)in bone marrow.They are considered to be the first cells to encounter antigen in peripheral tissues.They are the immune cells with the strongest antigen presenting effect at present.DCs can quickly recognize antibodies in infected sites and infiltrated pathological tissues,secrete cytokines and chemokines for migration,activate immature T cells,and promote T cell maturation and differentiation,participate in specific immune response.At present,the research of DCs in the field of cancer has made some progress,but there is still a lack of research on the disease progress in syphilis.Therefore,we tried to study the content of DCs and its subsets pDC and mDC in peripheral blood infected with Treponema pallidum(TP)and the expression difference of CD molecule by flow cytometry,so as to explore the influence of these changes and differences on the progress of TP infection,and seek a new way of early prevention,early diagnosis and effective treatment.Methods According to the exclusion and inclusion criteria,peripheral blood samples from 40 normal controls,37 patients with latent syphilis,46 patients with serum fixed syphilis,20 patients with secondary syphilis and 11 patients with neurosyphilis were collected.Mononuclear cells were extracted.The content of DCs and its subsets p-dc and m-dc and the expression of CD molecules were detected by flow cytometry,To explore the changes of DCs and its subsets in the process of TP infection.Result 1.A total of 40 normal healthy controls,16 males and 24 females,aged 41.70±9.97 years.There was no significant difference in age and gender between the groups.2.The expression of DCs was significantly increased in syphilitic patients with sero-resistance,patients with secondary syphilis and patients with neurosyphilis.3.There were significant differences in the expression levels of DCs subsets pDC cells among normal control group,latent syphilis,syphilitic patients with sero-resistance,secondary syphilis and neurosyphilis;There was no significant change in the expression level of mDC cells between groups4.Spearman correlation analysis showed that DCs and its subsets mDC and pDC cells in syphilis patients at all stages were correlated with different parameters of blood routine,coagulation function and biochemistry.Conclusion First,the expression level of DCs surface molecules in peripheral blood of patients with syphilis at all stages was significantly higher than that of normal people,indicating that stimulating the proliferation of DCs plays a relevant role in TP infection.Second,the expression level of pDC cell surface molecules in peripheral blood of patients with syphilis at all stages was significantly lower than that of normal people,and there was no significant change in the expression of mDC cells,suggesting that TP infects human body and induces the differentiation and maturation of DCs.pDC cells play a major role in this process.Third,DCs and its subsets pDC and mDC cells in each group are partially correlated with blood routine,coagulation function and biochemical parameters,but they are not universal. |
