Objective:To systematically evaluate the correlation between the curative effect of traditional Chinese medicine in the treatment of liver cancer and VEGF,and to explore the mechanism of "anti-tumor prescription" inhibiting angiogenesis of liver cancer.Methods:Meta-analysis was used to screen and analyze the clinical treatment literature of liver cancer in each database in the past 20 years,to analyze the correlation between the clinical effect evaluation of TCM treatment of liver cancer and VEGF.In this study,human hepatoma cells(HEPG2)were cultured with mouse drug-containing serum of traditional Chinese medicine "Anti-tumor Prescription",and the culture recovery liquid was collected for continued culture of human umbilical cord venous blood endothelial cells(HUVEC).After culture,corresponding samples were subjected to CCK-8,cell scratch test and HOE apoptosis staining test.The expression of VEGFR was detected by ELISA and WB..Results: Literature analysis results: TCM combined with Western medicine or chemotherapy can inhibit the generation of VEGF factors closely related to angiogenesis in tumor to a certain extent,and its total effective rate on liver cancer has a certain correlation with this inhibitory effect.Results:(1)CCK-8 results showed that the serum containing "Anti-tumor prescription" had inhibitory effect on the proliferation of Hep G2 cells,and the inhibitory effect was concentration-dependent.HUVEC cell proliferation was not significantly inhibited.The recovered culture solution of drug-containing serum cultured by Hep G2 had a relative inhibitory effect on the proliferation of HUVEC cells,and the inhibitory intensity was concentration-dependent.(2)Cell scratch test results showed that the serum containing "Anti-tumor prescription" could effectively reduce the migration ability of Hep G2 cells,but its recovery solution had no significant effect on the migration ability of HUVEC;(3)HOE staining results showed that the serum containing "anti-tumor prescription" could promote apoptosis of Hep G2 cells;However,it was not observed that the recovery solution could promote the apoptosis of HUVEC cells.(4)ELISA results showed that the serum containing anti-tumor formula could effectively reduce the secretion of VEGF in Hep G2 cells;(5)Western blot results showed that there was no significant effect of "Anti-tumor formula" containing serum on the expression of VEFGR in HUVEC cells after Hep G2 culture.Conclusion:(1)The combination of Traditional Chinese medicine with western medicine or chemotherapy can inhibit the production of VEGF,which is closely related to angiogenesis,in tumor to a certain extent,and its total effective rate on liver cancer may be correlated with this inhibitory effect(2)The serum containing "Anti-tumor prescription" can effectively inhibit the proliferation of HEPG2 cells,promote the apoptosis of HEPG2 cells and reduce their migration ability;(3)The drug-containing serum of "Anti-tumor prescription" has no direct inhibitory effect on the proliferation of HUVEC cells,and its relative inhibitory effect on the proliferation of HUVEC cells in the HEPG2 microenvironment may be realized by reducing the VEGF secretion of HEPG2 cells. |
