Cell adhesion and migration are common and important physiological processes in living organisms,and play significant roles in many physiological functions and pathological processes,such as embryo development,wound healing,immune response and tumor metastasis.Therefore,the regulation of cell adhesion and migration lays the groundwork for exploring the mechanism of tissue repair and regeneration,and it is of great significance for the design of scaffold materials.Hyaluronic acid(HA),as a natural polysaccharide,is widely found in the extracellular matrix of animals.It has excellent biocompatibility and biological function,and is widely used in the construction of biomaterials.In this thesis,the gradient surface and patterned surface of HA were fabricated via surface modification.By co-culturing of cells on the functionalized surfaces,the purpose of regulating cell behavior was realized,and the effects of HA gradient and pattern on cell adhesion and migration behavior were mainly explored.In Chapter 2,HA is immobilized onto the glass surface by amidation reaction.The density of HA is controlled by varying the reaction time at different positions of the glass surface.The gradient surface of HA is successfully fabricated.Material surfaces are characterized by water contact angle,surface potential,and fluorescence intensity.Then,the adhesion and migration behaviors of L929 and A549 are investigated by culturing the cells on the modified surface.The spreading area decreases with the increasing of HA surface density.The gradient of HA can induce directed migration of L929,but has no significant effect on directed migration of A549.The expression of vinculin,fibronectin and F-actin was analyzed by immunofluorescence staining,and the mechanisms of cell adhesion and migration are studied.In Chapter 3,in order to further study the effect of HA functionalized surface on cell behavior,the patterned surface of hyaluronic acid stripes is fabricated by oxygen plasma etching with a covered PDMS stamp.L929 and A549 cells are cultured on the patterned surface.Both L929 and A549 cells preferentially adhered to the plasma etched region with good orientation.The distribution and expression of vinculin,fibronectin and F-actin are also observed and analyzed by immunofluorescence staining.This thesis aims to reveal the mechanisms of HA functionalized surface in regulating the adhesion and migration of different type of mammalian cells.The results reported in this thesis may set a new starting point for the understanding of the interactions between natural polysaccharides and cells.
