Objective:To explore the impact of VEGF165 on neural function and the vessels around the hematoma in rat with cerebral hemorrhage model by injecting autologous caudal artery.Methods:60 male SD rats were randomly divided into sham-operated group,control group,VEGF165 group.There were 24 SD rats in control group and VEGF165 group,There were 12 SD rats in sham-operated group.Autologous tail arterial blood 50μl injection the caudate nucleus to make cerebral hemorrhage model.The sham-operated groupwas same as the rest two groups but injection blood.After cerebral hemorrhage model made in rats,adopt the NSS scores,the balance beam walking experiments and forelimbs placed experiment of judgment nerve function change.at 24 h,VEGF165 group was given 10 μl,the concentration is 0.025μg/μl into lateral ventricle.control group was given the same amount of saline injections of lateral ventricle,sham-operated group is not given intervention.Every group was divided into four subgroups,each subgroup had 6 rats in control group and VEGF165 group,sham-operated group only 3 rats in every subgroup.In 1 d,2 d,5 d,7 d,10 d gives the evaltation of neural the put to death in the rats and get the brain tissue.Different time of brain tissue detect CD34 protein expression through immunohistochemical staining and wesern-blot,adopting qPCR detect m RNA expression of MAP2.Results:In 2d,5d,7d,10 d control group NSS scores were 2.96±0.75,2.94±0.64,2.33±0.49,1.83±0.75;VEGF165 group were 2.25±0.79,1.33±0.49,1.00±0.43,0.50±0.55,compared two groups of different time(P < 0.05,the difference was statistically significant).The balance beam walking test results indicate the control group compared with VEGF165 group at different time points(P > 0.05,there was no statistically significant difference).Fore placement test result is shown in 2 d,5 d,7 d,10 d control scores were58.13±13.73,52.22±13.09,54.17±14.43,56.67±16.33;VEGF165 group were 70.83±14.72,78.89±11.83,76.67±22.29,90.00±10.95,compared two groups of different time(P < 0.05,the difference was statistically significant).HE staining results showed that compared to the 10 d and 5d the blood vessels in VEGF165 group were better than control group.Immunohistochemical staining results suggest in 2 d,5 d,7 d,10 d the number of cells expressd CD34 is 438±45,569±105,732±90,903±103in control group;the number of cells expressd CD34 is 1506±179、1607±180、1725±92、2186±212 in VEGF165 group,compared two groups of different time(P < 0.05,the difference was statistically significant).According to the Western blot results in 2 d,5 d,7 d,10 d control group compared with VEGF165,the relative expression of CD34 was lower than that in group VEGF165.qPCR results in 2 d,5 d,7 d,10 d the MAPA relative expression were 0.10±0.27,0.07±0.00,0.32±0.10,0.59±0.19 in control group;VEGF165 group were 0.65±0.15,1.34±0.93,2.09±0.60,2.29±0.77;At different times compared the two groups(P < 0.05,the difference was statistically significant).Conclusions: In early cerebral hemorrhage application of VEGF165 is beneficial to the recovery of neural function,to improve the reaction ability after hemorrhage in rats.Cerebral hemorrhage early VEGF165 could promote the regeneration of the neural dendrites,can promote the regeneration of the cerebral hemorrhage haematoma around blood vessels.
