Objectives The physical and chemical properties of tantalum-containing new titanium alloys prepared by powder metallurgy sintering were analyzed.Through in vitro experiments,the biological effects of new titanium alloys on preosteoblasts and osteogenic differentiation were evaluated at the cellular and molecular levels,to provide experimental basis for the development of new materials.Methods Experiment 1.Physicochemical properties of new titanium alloy containing tantalum:The experiment was divided into five groups,namely,A:Ti-10Mo-28Nb-3Zr-2Ta,B:Ti-10Mo-28Nb-3Zr-4Ta,C:Ti-10Mo-28Nb-3Zr-6Ta,D:Ti-10Mo-28Nb-3Zr-8Ta,E:Ti-6Al-4V.ABCD was the experimental group,and E was set as the control group.The surface morphology,elemental composition,roughness and hydrophilicity of the specimen were detected.Experiment 2.Effect of new titanium alloy containing tantalum on osteogenic properties of cells:The experimental groups were the same as above.The effects of five groups of specimens on cell adhesion and morphology,alkaline phosphatase activity expression,osteocalcin and type I collagen gene expression and mineralization nodule formation were compared.All data were analyzed by statistical software SPSS 24.0,and P<0.05 was set to indicate the difference was statistically significant.Analysis of physical and chemical properties of new titanium alloy.The effect of new titanium alloy on cell growth and osteogenic differentiation was evaluated.Results 1 Physicochemical properties of the new titanium alloy containing tantalum:the specimens all contained Ti,Mo,Nb,Zr and Ta components.Under SEM,the pore structure beneficial to cell adhesion and growth was observed on the surface of the specimens.The surface roughness values of the five groups of specimens were 3.18±0.06μm,3.26±0.10μm,3.21±0.10μm,3.25±0.12μm,and 3.17±0.04μm,respectively,with no statistical difference(P>0.05).The contact angles of the five groups of specimens were 56.68±0.81°,57.17±0.69°,55.59±0.47°,56.51±0.74°,and62.80±0.71°,respectively,with statistical difference(P<0.05)the experimental group and the control group(P<0.05),which provided a favorable environment for after implantation.2 Bone promoting effect of new titanium alloy containing tantalum:In the early stage,five groups of specimens had no significant effect on cell adhesion(P>0.05);The effect of specimen on the ALP activity of cells:on the 4th day,the experimental data of the five groups were 0.064±0.001,0.189±0.032,0.106±0.013,0.097±0.001 and 0.056±0.001.The ALP activity of group B was the highest,and the difference was statistically significant(P<0.05).On the seventh day,the experimental data of the five groups were 0.137±0.042,0.528±0.165,0.285±0.034,0.250±0.070and 0.077±0.010.The value of group B was the highest and the difference was statistically significant(P<0.05).Observation of mineralized nodules and detection of Ca2+concentration:the mineralized nodules in group B were the most formed and the staining was the deepest.Compared with Ca2+concentration(1.20±0.06,1.32±0.08,1.20±0.02,1.21±0.05,1.16±0.08),it can be found that the value of group B was the highest and the difference was statistically significant(P<0.05).The expression of OCN gene and COL-I gene:On the seventh day,the expression of the two osteogenic genes was detected,and the expression of the two groups of genes was the highest in the cells cultured extract of group B,and the difference was statistically significant(P<0.05).Considering multiple experimental results,the osteogenic effect of Ti-10Mo-28Nb-3Zr-4Ta group was better than that of the other four groups,and the difference was statistically significant.Conclusions 1 The surface micromorphology of Ti-10Mo-28Nb-3Zr-xTa new titanium alloy prepared by powder metallurgy has a promoting effect on cell adhesion and growth.There was no significant difference in the surface roughness of the five groups of specimens,and they were all within the range of suitable osteoblast adhesion and growth;The contact angles of the five experimental groups were all less than 90°and were different from those of the control group,indicating that the hydrophilicity of the new material was better than that of the traditional medical titanium alloy.2 Experiments showed that the new Ti-10Mo-28Nb-3Zr-4Ta titanium alloy had the best effect on promoting bone formation.Figure 9;Table 17;Reference 104...
