| Objective:SOX protein expression and its function presented a close relations in proliferation and apoptosis of human lung cancer cells.This paper aimed to study the SOX9 expression in human lung tissue.in order to provide new train thoughts and experimental basis for further research of tumor molecu Lar mechanisms and gene therapy on lung cancer cells.Methods:Take the human lung cancer cell line A549,then setting the control group and research group.Control group did not take any processes,and take the SOX9 antibodies into the tumor tissue of test group.Then detect the specific expression of SOX9 by semi-quantitative reverse transcription PCR and fluorescence quantitative PCR methods.Raising the SOX9 concentration in lung cancer cells,by the fluorescein experimental analysis to analyze the m RNA transcription activity of Wnt1 and β-catenin,as well as the transcription activity of β-catenin/TCF.By the Western-Bolt method to test the expression level of Wnt1,β-catenin,Cyclin D of the Wnt/β-catenin signaling pathway in lung cancer cells.Using the immune confocal microscopy method to detect the nuclear transfer of β-catenin.By the MTT method to detect the cell proliferation and apoptosis after the implantation of SOX9,do the later data statistics and analysis.Resu Lt:Research showed that the SOX9 cou Ld get clearly express in test group in comparison with the control group,statistical data showed that the P value was less than 0.05 in 24 hours and 48 hours,within statistically significant difference.SOX9 expressions cou Ld significantly inhibit the Wnt1 and β-catenin m RNA transcription activity in lung cancer cell.Increasing the concentration of SOX9,the inhibitory increased significantly.The key proteins expression level of Wnt1,β-catenin and Cyclin D dropped significantly at the same time.The nuclear transfer of β-catenin also obviously impeded.Through the effective expression of SOX9 in lung cancer cells,the tumor cell proliferation level declined obviously,and meanwhile the tumor cell apoptosis increased.Conclusion:SOX9 is the important lung tumor suppressor genes.SOX9 do the directly negative regu Lation by the Wnt/β-catenin signaling pathway.SOX9 can fu Lly inhibit the lung tumor cell proliferation and inducing apoptosis process. |
