| The role of transcription factors in cancer has attracted significant attention.Mist1,a component of BHLH transcription factors,has been documented to be an important factor in the tumorigenicity and tumor progression of pancreatic ductal adenocarcinoma.We clarified that Mist1 could bind to the promoter of COL11A1 as a transcription factor and repress its transcription by the Ch IP and dual-luciferase assays.Meanwhile,we found that the N-terminal repressor region of Mist1 was capable of inhibiting COL11A1 expression.Analysis of the GEPIA(Gene Expression Profiling Interactive Analysis)web tool showed that the expression of COL11A1 was upregulated in PDAC and positively correlated with a worse prognosis for patients with PDAC.Knockdown of COL11A1 inhibited the proliferation and invasion of PDAC cell lines,while upregulating COL11A1 increased those abilities.Overexpression of COL11A1 could apparently reverse the inhibition of pancreatic cancer cell migration and proliferation by overexpression of Mist1.These results demonstrated COL11A1 as a carcinogen in PDAC,and it acts as the key target gene of Mist1 on tumor progression of PDAC. |
