| According to domestic and foreign investigations,long-term surviving activities in space can pose potential hazards to the reproductive functions of male and female astronauts.However,the protection measures for reproductive system damage caused by simulated weightlessness environment still need to be further studied,and the screening of specific drugs still needs the support of a large amount of experimental data.In this experiment,the crude polysaccharides of Atractylodes macrocephala extracted from the traditional Chinese medicine Atractylodes macrocephala were used as experimental materials,and the rats were gavaged at a fixed time every day for 30 days.The rats were divided into 5 groups,namely the ground blank control group and the simulated micro Gravity group,low-dose gavage group,medium-dose gavage group,high-dose gavage group.After 30 days of tail hanging treatment,complete the collection within two days,and conduct morphological and molecular biological research on the testicular tissue.The histological level of the testis is studied by paraffin section and HE staining methods,and screened according to the changes in morphology and tissue.The effective concentration of Atractylodes macrocephala polysaccharide to alleviate the testis damage of male rats caused by microgravity environment.At the level of molecular biology,ELISA kits were used to detect changes in serum levels of sex hormones.The results showed that after simulated microgravity treatment,the morphology and organization of the rat testis changed significantly,and the serum sex hormone content changed significantly,while the serum sex hormone content index of the rats in the Atractylodes macrocephala polysaccharide administration group returned to the normal level,and the tissue morphology was improved.The experimental results of molecular biology showed that the rat testis tissue apoptosis,autophagy,and antioxidant-related genes changed significantly after tail hanging,while the gene expression of rats after intragastric administration of Atractylodes macrocephala was compared with the simulated microgravity group.There is a more obvious improvement.In summary,the crude polysaccharides of Atractylodes macrocephala play a certain degree of protection against damage to the reproductive system of rats caused by simulated microgravity environment,alleviate the changes in the content of sex hormones caused by reproductive damage,and regulate genes caused by the microgravity environment.However,the optimal concentration and the mechanism of Atractylodes macrocephala polysaccharides to prevent reproductive damage need to be further studied. |
