| Histone post-translational modification plays a key role in the regulation of gene expression and chromatin state,which enhances its research value in physiological and pathological state.With the growth of histone modification data,this project updated the human histone database(HHMD).In this study,we developed HHMD version2.0 database,which manually collected 833 relationship information of 17 histone modifications in 9 cancer States,including 57 histone modifications based on ChIP-seq technology.The number of normal and diseased tissue / cell samples reached 207 and 351 respectively(diseased tissue 23,cell line 328).At the same time,123 sets of Hi-C data and 96 sets of ATAC-seq sequencing data were included.At the same time,it integrates the interaction information of drugs,transcription factors and target genes,The data of ChIP-seq,Hi-C and ATAC-seq were processed through a unified standardized analysis process,and then the histone modification information,chromatin interaction information and chromatin open state information were determined.In order to further explore the driving role of histone modification in diseases,HHMD version2.0 screened out the characteristics of different histone modifications in 25 diseases.Based on the above data,this project draws the network of histone modification gene disease / cell line / normal tissue respectively,which directly shows the relationship between histone modification and sample type.According to the histone modification characteristics identified in disease tissues /cell lines,combined with the drug target database,we also constructed the histone modification gene target network.At the same time,by integrating Hi-C and ATAC-seq data,the information of chromatin opening and interaction in histone modified regions was increased.In addition,in order to analyze the regulatory function of histone modification,HHMD version2.0 provides genome visualization function to comprehensively display histone modification,transcription factor target gene,super enhancer,Cp G island and gene mutation.In conclusion,the goal of HHMD version2.0 is to develop a comprehensive landscape of histone modification,which can be used to reveal the regulatory function of histone modification and the research of disease treatment targets. |
