Capsaicin,(trans)-8-methyl-N-vanillyl-6-nonenamide,is a flavonoid isolated from Capsicum annuum L.,a well-known plant.It can interact with the TRPV1 receptor to make human body feel pain.It has been demonstrated that capsaicin possesses various biological activities,including anti-inflammatory,anti-tumor,anti-bacteria and e.t.c.NLRP3 inflammasome plays an important role in many inflammatory diseases in The contexts of infections.Although capsaicin has been reported to inhibite inflammatory diseases,while it is still unclear the affect of capsaicin during NLRP3 inflammasome activation and the underlying action mechanism.Here we show that capsaicin specifically inhibits NLRP3 inflammasome activation,as well as the secretion of mature IL-1β,K~+efflux and the ROS.Furthermore,we discover the therapeutic effect of capsaicin in sepsis of mice.During NLRP3 inflammasome activation,BMDMs secretes inflammatory cytokines,such as IL-1βand caspase-1,which leads to the organic damage and a sudden shock.We aimed to research the role of capsaicin in NLRP3 inflammasome activation and the preliminary mechanism,as well as the potential value in the treatment of NLRP3-related inflammaory disteases. |
