| Objective: Non-codingRNA plays an increasingly important role in the prediction of susceptibility genes and small molecule drugs for gestational hypertension.By studying the differential LncRNA and mRNA expression profiles of placental tissues in patients with hypertension during pregnancy,the mechanism of action of LncRNA and its target genes is explored to provide a theoretical basis for clinical diagnosis and treatment.Methods: Thirty patients with pregnancy-induced hypertension who were treated in third-grade class-A hospital of Weifang from May 2018 to May 2019 and delivered at full term were selected as the experiment group,and thirty pregnant women who were in the hospital’s healthy obstetric examination and delivered at full term during the same period were selected as the control group.Two sets of placental tissues were taken forRNA sequencing;the differential LncRNA and mRNA were screened byRNA-seq,and the target genes of differential LncRNA were predicted using the database;GO and KEGG analysis was performed using cluster profiler to enrich the biological function and enrichment of the target gene respectively Integrated signaling pathway;using STRING online database and Cytoscape software for protein-protein interaction network construction,LncRNA-miRNA-mRNA network and differentially expressed mRNA-small molecule drug association prediction analysis.Results: Through analysis of the two sets of data,19 differential hypertension-related Lnc RN(4 up-regulated LncRNA and 15 down-regulated LncRNA)were obtained.There were 423 differentially expressed mRNAs(84up-regulated mRNA and 339 down-regulated mRNA)(P < 0.05);through the enrichment analysis of GO and KEGG,it was found that pregnancy-induced hypertension is related to endothelial cell damage,inflammatory response,abnormal immune regulation,abnormal trophoblast invasion,etc.By constructing PPI network and LncRNA-miRNA-mRNA network,2 Significantly enriched sub-network modules:module-A(score = 15)contains 15 nodes and 105 interaction pairs;module-B(score= 10)contains 17 nodes and 117 interaction pairs,and Linc01128 is on the network The most regulated miRNAs and mRNAs have the core role in the network;the differentially expressed gene-drug small molecule prediction results found that there are 19 pairs of differential genes corresponding to small molecule drugs,including antibodies and inhibitors,antagonists and other relationships.Conclusion: Differentially expressed LncRNA in the placenta of patients with pregnancy-induced hypertension can participate in the regulation of various biological function-related signaling pathways through targeted regulation of their target genes,and play an important role in the development and development of pregnancy-induced hypertension.Small molecule drugs can be used as clinical reference basis for treatment. |
