Objective:The purpose of this study was to evaluate the antibacterial effect of Cangai Volatile Oil(CAVO)on potential pathogenic bacteria of children’s hand skin,in order to provide some experimental basis for the development of convenient,green and natural Chinese herbal compound Volatile Oil skin disinfectant,and to provide reference for the interpretation of the scientific connotation of the theory of“aromatic herbe can oppress the turbid”in Chinese medicine.Methods:1.Detection of potential pathogenic bacteria on children’s hand skinUnder aseptic operation,100 children’s hand skin samples were collected from the pediatric outpatient department of Traditional Chinese Medicine Hospital of Yunnan.The samples were detected by 16S RNA high-throughput sequencing technology,and the potential pathogenic bacteria of children’s hand skin were analyzed.2.Identification test of removal effect of residual disinfectantThe samples of the tested bacteria treated with CAVO were diluted and then filtered and rinsed to determine whether the residual CAVO could be physically removed.3.The killing of potential pathogenic bacteria in children’s hand skin by CAVOSuspension quantitative germicidal test and filtration method were used to study the killing effect of CAVO on children’s hand skin potential pathogenic bacteria.4.Minimum inhibitory concentration(MIC)testUsing the filter paper method,different concentrations of CAVO were added to the filter paper,and the filter paper was pasted on the solid medium coated with the suspension of bacteria.The MIC of CAVO against potential pathogenic bacteria on children’s hand was determined according to the size of the bacteriostatic circle.Results:1.Detection of potential pathogenic bacteria on children’s hand skinA total of 406145 effective sequences and 336727 optimized sequences were obtained from 100 samples of children skin microorganisms by high-channel sequencing,and the average sequencing length was between 412 and 418bp.At 97%similarity level,the number of effective OTUs was 1881,and 28 phyla,59 classes,153 orders,292 families and 662 genera were identified in the community composition.At the level of bacteria,Staphylococcus,Escherichia and Streptococcus with potential pathogenic bacteria were detected.2.Identification test of removal effect of residual disinfectantUnder the condition of 20±1℃,Staphylococcus aureus(Staphylococcus aureus,S.aureus)was used as the test strain,which was divided into the following groups:(1)the first group:suspension culture of CAVO+bacteria to observe the killing or inhibitory effect of CAVO on the tested bacteria.The second group:(bacterial suspension+CAVO)+filter washing method to observe whether the drug removal treatment can revive the growth of the tested bacteria.The third group:(bacterial suspension+water)+filtration washing method to observe the effect of drug removal treatment on the growth of the tested bacteria.The fourth group:bacterial suspension culture,which was the positive control group.The experiment was repeated three times,and the results showed that the average number of growth bacteria in group 1 and group 2 was 0CFU/m L and 200 CFU/m L, respectively,the average number of bacteria in group 3 was 2.40×10~4CFU/m L,the average growth in group 4 was 2.90×10~4CFU/m L,and the average error between groups3 and 4 was 8.99%.3.The killing of potential pathogenic bacteria in children’s hand skin by CAVOThe test was repeated three times at 20±1℃.The results showed that the potential pathogenic bacteria of children’s hand skin caused by CAVO original solution were common pathogenic bacteria:The average bactericide rates of Staphylococcus aureus(S.aureus),Escherichia coli(E.coli)and Group A beta-hemolytic Streptococcus(GAS),after 1min treatment were 99.80%,99.26%and 99.77%,respectively.4.Minimum inhibitory concentration(MIC)testCAVO has a MIC of 25%for S.aureus,50%for E.coli,and 50%for GAS.Conclusion:1.Children’s skin microbes detected potential pathogenic bacteria as Staphylococcu,Escherichia,and Streptococcus,suggesting that S.aureus,E.coli,and GAS may be common pathogenic bacteria on children’s hand skin.2.CAVO has an inhibitory effect on the potential pathogenic bacterium(S.aureus,E.coli,GAS)on children’s hand skin,and has a dose-effect relationship,which providing experimental basis for the development of CAVO children’s skin disinfectant.
