Objective: Through the detection of Nanog and Snail in ovarian serous carcinoma tissues,we analyze the expression of Nanog and Snail in ovarian plasmacytoma tissues and their relationship with various parameters of patients’ clinicopathology,and explore the role and clinical significance of Nanog and Snail in the occurrence,development and infiltration and metastasis of ovarian plasmacytoma,and evaluate the guiding significance of Nanog and Snail in the diagnosis of ovarian plasmacytoma.We study the application of immunohistochemical methods to detect the expression characteristics of Nanog and Snail in ovarian plasmacytoma tissues,analyze the correlation existing between them and the clinicopathological parameters of patients through their expression,and explore the changes of Nanog and Snail during the development,progression and infiltration of ovarian plasmacytoma and their clinical significance,so as to provide a new way to study the origin and biological behavior of tumors,and provide a new approach to the treatment of tumors,bring new methods for the treatment of tumors and bring gospel to tumor patients.Methods:A total of 100 ovarian histopathological paraffin specimens were collected from October 2016 to October 2019 from patients who underwent surgical treatment at the Department of Gynecology,People’s Hospital of Mengyin County.All specimens from the enrolled patients were checked by 2 pathologists.All patients were non-pregnant,free of autoimmune diseases,and did not receive any radiotherapy,chemotherapy and other immunological treatments before surgery and gave their informed consent.The ovarian tissues of all patients were divided into three groups according to the pathological results,namely,control group,ovarian junctional plasmacytoma and ovarian plasmacytoma group.There were 20 cases in the control group,20 cases in the ovarian junctional plasmacytoma group,and 60 cases in the ovarian plasmacytoma group.Firstly,the positive expression rates of Nanog and Snail genes were detected in the three different groups respectively.Secondly,for the 60 cases of ovarian plasmacytoma,the positive expression rates of Nanog and Snail genes were compared longitudinally within the groups according to different clinical stages and whether lymphatic metastases were present.The correlation between Nanog and Snail was also analyzed.The statistical software was SPSS23.0.Results:1.Among 20 normal ovarian tissues,a total of 3 cases were positive for Nanog expression,with a positive rate of 15%;among 20 ovarian junctional plasmacytoma tissues,a total of 12 cases were positive for Nanog expression,with a positive rate of60.0%;among 60 ovarian plasmacytoma tissues,a total of 49 cases were positive for Nanog expression,with a positive rate of 81.67%.From the above results,it can be seen that the positive rate of Nanog expression gradually increased from normal ovarian tissues to ovarian plasmacytoma tissues,and the difference of positive Nanog expression in the three different tissues was statistically significant,with the lowest positive Nanog expression in normal ovarian tissues and the highest positive Nanog expression in ovarian plasmacytoma tissues,and the difference was statistically significant when comparing the two.2.Among 21 cases of low-grade lesioned ovarian plasmacytoma tissues,a total of 12 cases were positive for Nanog expression,with a positive rate of 57.14%;among 39 cases of high-grade lesioned ovarian plasmacytoma tissues,a total of 37 cases were positive for Nanog expression,with a positive rate of 94.87%.From the above results,it can be seen that the positive rate of Nanog expression gradually increased from low-grade lesioned ovarian plasmacytoma tissues to high-grade lesioned ovarian plasmacytoma tissues,and there was a statistically significant difference in the expression of positive Nanog expression in different tissues,2=12.978,P=0.000.3.Among 27 tissues with clinical stage I-II ovarian plasmacytoma,a total of 19 cases were positive for Nanog expression,with a positive rate of 70.37%;among 33 tissues with clinical stage III-IV ovarian plasmacytoma,a total of 30 cases were positive for Nanog expression,with a positive rate of 90.91%.The positive expression of Nanog in clinical stage III-IV ovarian plasmacytoma tissues was higher than that in clinical stage I-II ovarian plasmacytoma tissues,and the difference was statistically significant.4.Among the 31 ovarian plasmacytoma tissues with lymph node metastasis,a total of29 cases had positive Nanog expression,with a positive rate of 93.55%;among the 29 ovarian plasmacytoma tissues without lymph node metastasis,a total of 20 cases had positive Nanog expression,with a positive rate of 68.97%.The positive expression of Nanog in ovarian plasmacytoma tissues with lymph node metastasis was higher than that in ovarian plasmacytoma tissues without lymph node metastasis,and the difference was statistically significant.5.Among the 20 normal ovarian tissues,there were 2 cases with positive Snail expression,and the percentage of positive expression was 10%;among the 20 ovarian junctional plasmacytoma tissues,there were 11 cases with positive Snail expression,and the percentage of positive expression was 55%;among the 60 ovarian plasmacytoma tissues,there were 47 cases with positive Snail expression,and the percentage of positive expression was 78.33%.From the above results,it can be seen that the positive rate of Snail expression gradually increased from normal ovarian tissues to ovarian plasmacytoma tissues,and the difference of positive Snail expression in the three different tissues was statistically significant,with the lowest positive Snail expression in normal ovarian tissues and the highest positive Snail expression in ovarian plasmacytoma tissues,and the difference was statistically significant when comparing the two.6.In this study,among 21 low-grade lesions of ovarian plasmacytoma tissues,a total of 11 cases were positive for Snail expression,with a positive rate of 52.38%;among 39high-grade lesions of ovarian plasmacytoma tissues,a total of 36 cases were positive for Snail expression,with a positive rate of 92.31%.From the above results,it can be seen that the positive rate of Snail expression gradually increased from low-grade lesioned ovarian plasmacytoma tissues to high-grade lesioned ovarian plasmacytoma tissues,and there was a statistically significant difference in the expression of positive Snail expression in different tissues,2=12.821,P=0.000.7.Among 27 tissues with clinical stage I-II ovarian plasmacytoma,a total of 18 cases were positive for Snail expression,with a positive rate of 66.67%;among 33 tissues with clinical stage III-IV ovarian plasmacytoma,a total of 29 cases were positive for Snail expression,with a positive rate of 87.88%.The positive expression of Snail in clinical stage III-IV ovarian plasmacytoma tissues was higher than that in clinical stage I-II ovarian plasmacytoma tissues,and the difference was statistically significant.8.Among the 31 ovarian plasmacytoma tissues with lymph node metastasis,a total of28 tissues were positive for Snail expression,with a positive rate of 90.32%;among the 29 ovarian plasmacytoma tissues without lymph node metastasis,a total of 19 tissues were positive for Snail expression,with a positive rate of 65.52%.The positive expression of Snail in ovarian plasmacytoma tissues with lymph node metastasis was higher than that in ovarian plasmacytoma tissues without lymph node metastasis,and the difference was statistically significant.9.Among them,there were 5 cases with positive expression of Nanog and negative expression of Snail,and 3 cases with positive expression of Snail and negative expression of Nanog;Spearman’s rank correlation analysis was performed(r=0.687,P=0.000).The results showed that the expression of Nanog and Snail in ovarian plasmacytoma tissues were positively correlated.Conclusion: 1.In ovarian plasmacytoma tissues,Nanog expression was higher than that in normal ovarian tissues,and Nanog expression was related to tumor grade,clinical stage,tumor malignancy and the presence of lymph node metastasis,and Nanog expression in ovarian plasmacytoma tissues with clinical stage III-IV was higher than that in ovarian plasmacytoma tissues with clinical stage I-II,and ovarian plasmacytoma tissues with lymph node metastasis were positively expressed.The expression of Nanog was higher in ovarian plasmacytoma tissues with lymph node metastasis than in ovarian plasmacytoma tissues without lymph node metastasis.2.In ovarian plasmacytoma tissues,Snail expression was higher than that in normal ovarian tissues,and Snail expression was related to tumor grade,clinical stage,tumor malignancy and the presence of lymph node metastasis,and Snail expression was higher in ovarian plasmacytoma tissues with clinical stage III-IV than in ovarian plasmacytoma tissues with clinical stage I-II,and in ovarian plasmacytoma tissues with lymph node metastasis.The positive expression of Snail was higher than that of ovarian plasmacytoma tissues without lymph node metastasis.3.The correlation between the expression of Nanog and Snail in ovarian plasmacytoma tissues was positively correlated by analysis.
