OBJECTIVE:In order to research the influence of different types of root canals on the apical sealing ability,we use glucose quantitative method to detect the apical microleakage after the different shapes of root canals was filled by three root canal filling methods,so as to reflect the apical sealing ability and provide reference for clinical practice.METHODS:Select freshly extracted premolars and molars, and use a low speed diamond disc to cut the teeth into 10 mm tooth roots for spare. We separately selected 30 regular straight root canals,oval-shaped root canals and moderate and severe curved root canals, which met the requirements by CBCT photography, then divided the three groups into three subgroups, namely A1、A2、A3,B1、B2、B3 and C1、C2、C3 groups, with 10 samples in each group. The negative control group(D group)and the positive control group(E group)each had 5 roots. After the preparation of the root canals by the Ni-Ti Pro Taper,Group A1,B1,and C1: iRoot SP + warm vertical condensation techniqued(WVCT);Group A2,B2,and C2: iRoot SP + cold lateral condensation technique(CLCT);Group A3,B3,and C3: iRoot SP + single-cone obturation technique(SCOT).When the root canal obturation of all samples were completed,we use the glucose microleakage detection model to detect the apical microleakage,so as to obtain the apical sealing performance of different types of root canals with three kinds of root canal obturation methods.RESULTS:Detect the apical micro-leakage in all samples by glucose quantitative method. There was no significant difference on the 1,7,14,21,and 28 day(P>0.05)for the 3 groups of regular straight root canals; There was no significant difference on day 1,7,and 14(P>0.05)for the 3 groups of moderate to severe curved root canals. But on day 21 and 28,the apical micro-leakage of CLCT group was significantly higher than WVCT group and SCOT group(P<0.05),and there was no significant difference between the latter two groups(P>0.05);There was no significant difference on the first day(P>0.05)for the 3groups of oval shaped root canals. But on day 7,14,21,and 28,the apical micro-leakage of CLCT group was significantly higher than WVCT group and SCOT group(P<0.05),and there was no significant difference of the latter two groups(P>0.05).CONCLUSION:In this paper,a new root canal filling material iRoot SP was selected as root canal sealer, and the microleakage of regular straight root canals, oval shaped root canals and moderate to severe curved root canals were detected after filling by three different root canal obturation methods. We have come to the following three conclusions.1.When filling regular straight root canals, there is no significant difference in apical microleakage among the three root canal obturation methods, they show similar apical sealing performance which are all good.2.When filling the oval-shape root canals, the results of CLCT group was higher than WVCT group and SCOT group at the beginning, the apical sealing performance was not good, and the latter two groups was better.3.When filling moderate and sere curved root canals, there were no difference between the results of three root canal obturation methods in short time, but the long-term effect of the CLCT group can not be guaranteed. The apical sealing performance of WVCT and SCOT was better than that of CLCT.
