Background:Bladder cancer is a common malignant tumor of urogenital system,which poses a serious threat to human health.Moreover,the incidence rate has increased in recent years.According to statistics,bladder cancer is the ninth most common cancer in the world,and the seventh most common male cancer in the world,incidence rate is about 4.7%of all cancers.According to statistics,there are about 430000 new cases in the whole year,and about 165000 patients died of bladder cancer every year.Male patients account for 75%of all new cases,but according to statistics,the mortality of female patients is higher.With the rapid development of molecular biology technology,many kinds of oncogenes related to the pathogenesis and progress of bladder cancer have been found,and we have a deeper understanding of the pathogenesis and treatment of bladder cancer.However,we still can not find an effective indicator to monitor the progress of bladder cancer,so we should do more research on molecular biology in the occurrence and progress of bladder cancer.In this study,we investigated the value of ska3 gene in the early diagnosis of bladder urothelial carcinoma.Objective:Spindle and kinetochore associated complex(ska)is a hot research topic at home and abroad recently.As a member of the family of ska complexes,the study of ska3 also attracts attention.Current studies suggest that ska3 gene is highly expressed in some malignant tumor cells,such as cervical cancer,breast cancer,hepatocellular carcinoma and prostate cancer.In this paper,we studied the expression of ska3 gene in normal bladder mucosa and bladder urothelial carcinoma with different clinicopathological stages,and elucidated the relationship between the expression of ska3 gene and age,gender,high risk factors and different clinicopathological stages of bladder carcinoma,so as to provide preliminary theoretical basis for early diagnosis and targeted therapy of bladder carcinoma.method:Immunohistochemistry and Western blot(SP)were used to detect the expression of ska3 protein in 52 paraffin embedded specimens of bladder urothelium and paracancerous tissues.The difference between the expression of ska3 protein in bladder cancer and paracancerous tissues was analyzed,and its relationship with gender,age,pathological grade,clinical stage and lymph node metastasis was analyzed.Then the experimental data were analyzed by SPSS 24.0 software,The measurement data are expressed as(χ±s)When p value is less than 0.05,it is considered that the difference is statistically significant.Result:1.The expression of ska3 in bladder urothelial carcinoma cells was significantly higher than that in paracancerous cells;2.Immunohistochemistry showed that the expression level of ska3 protein in bladder urothelial carcinoma was positively correlated with pathological stage,clinical grade and lymph node metastasis(P<0.05)3.The result of Western blot showed that the expression of ska3 protein in bladder urothelial carcinoma was(3.091 1±104),and the expression level in normal bladder tissue was(0.693±467),indicating that the expression of ska protein in bladder cancer tissue was significantly higher than that in normal bladder tissue,and the difference was statistically significant(P<0.05);Conclusion:the expression of ska in bladder urothelial carcinoma is related to the pathological stage,clinical grade and lymph node metastasis.
