Background Neuroblastoma(NB)is the most common extracranial solid cancer in children.Most of the children can die rapidly,which seriously affects the survival of children.NB has obvious heterogeneity,and its clinical manifestations are very different,and its treatment is also based on comprehensive treatment.At present,the treatment of NB is constantly being explored in the world.Studies have found that the Notch1 signaling pathway is related to the occurrence and development of neuroblastoma.Timosaponin AⅢ(TA Ⅲ)is a traditional Chinese medicine that can inhibite a variety of tumors,but there is no relevant report on whether Timosaponin AⅢ can affect the NB and Notch1 signaling pathway.Objective This study mainly explored the effects of Timosaponin AⅢ on cell proliferation and apoptosis in NB cell line SK-N-BE(2)cells and its mechanism,and provided new treatment methods for the treatment of NB.Methods Different concentrations of Timosaponin AⅢ was acted on SK-N-BE(2)cells.The changes of cell morphology were observed under an inverted microscope.The cell proliferation was detected by Cell Counting Kit-8 assay(CCK-8).Used flow cytometry analyse the cell apoptosis.The expression of Notch1 signaling pathway related target genes Notch1,c-Myc and Hes1 mRNA was detected by Reverse transcription-quantitative polymerase chain reaction(RT-q PCR).The western blot was used to detect the expression of the related target protein c-Myc and Hes1.Results The experiment found that Timosaponin AⅢ could effectively inhibit proliferation in SK-N-BE(2)cells in vitro,and the inhibitory effect was characterized by concentration and time dependence.Simultaneously,Timosaponin AⅢ also promoted the apoptosis of SK-N-BE(2)cells.Moreover,Timosaponin AⅢ decreased the expression levels of Notch1 signaling pathway-related target genes and target proteins(Notch1,c-Myc and Hes1)in SK-N-BE(2)cells.Conclusion Timosaponin AⅢ may exerts anti-tumor effects by inhibiting the Notch1 signaling pathway in neuroblastoma SK-N-BE(2)cells.Our study indicate that Timosaponin AⅢ can be a new option for the treatment of NB. |
