Objective:Changping decoction(CPD)is an effective prescription for the treatment of diarrheal irritable bowel syndrome(IBS).In this study,animal experiments were used to observe the efficacy of CPD in the treatment of IBS,and combined with literature review and bioinformatics methods to explore the mechanism of CPD for IBS.Methods:The 32 Wistar rats were divided into blank group,model group,treatment group and control group.The IBS rat model was established through water avoidance stress experiment.The treatment group was given CPD continuous gavage for 14 days,and the control group was given normal saline gavage for 14 days.After the treatment,we observe the rat’s fur condition,mental state,weight change and stool morphology.Using the colorectal dilatation method to detect the visceral pain threshold between different groups.Using pathological observation to compare the pathological changes of colon tissue between different groups.Using immunohistochemistry for central nervous system Targets and the expression of related targets on the brain-gut axis is detected.Screening the possible targets of CPD by consulting literature and bioinformatics methods.Using immunization Histochemistry detects the expression of core targets and brain-gut axis-related targets.Results:The rats in the blank group and the treatment group have neat and shiny fur,sensitive response,good mental state,more weight gain,and granular feces.The rats in the model group and the control group showed loose fur and dim color,lethargy,reduced activity,fatigue,curled up,arched back,drooping beard,little or no weight gain,and loose stools and thin perianal body fur were contaminated,suggesting that the symptoms of treatment group IBS rats have been relieved.It is found that the visceral pain threshold of the treatment group and the blank group is significantly higher than that of the model group and the control treatment group through the colorectal expansion method,suggesting that the visceral hypersensitivity of treatment group IBS rats has been improved.Histopathological observation found that the colon tissue of the model group and the control group showed the phenomenon of lymphocyte proliferation,but the blank group and the treatment group did not,suggesting that the intestinal inflammation of IBS rats has been improved.Literature review and bioinformatics analysis suggested that VEGFA,MAPK8,PTGS2,CRH and HTR2A may be the targets of CPD for IBS.The results of immunohistochemistry showed that the expression of related targets in the model group and the control group was higher than that in the blank group and the treatment group.It was found that the CPD could inhibit the expression level of related targets.Conclusion:The CPD can relieve the symptoms of IBS rats,reduced the visceral hypersensitivity of IBS rats,improved intestinal inflammation.The immunohistochemical results suggest that CPD improved intestinal inflammation by down-regulating the expression levels of MAPK8,VEGFA and PTGS2,and CPD inhibited the activation of the HPA axis via downregulating the expression levels of CRH and HTR2A.
