Research background: oral squamous cell carcinoma(OSCC)is the common malignance in head and neck,and the five-year survive rate of these patients remained approximately50 percentages,though great advances achieved in therapies.And the prognosis of advanced OSCC patients is required to be improved.Chemotherapy is the important treatment for OSCC patients,which including platinum,fluorouracil,paclitaxel and methotrexate etc.However,theses traditional chemotherapeutic agents are often restricted by drug resistance and adverse reactions such as myelosuppression,gastrointestinal reaction,mucositis,Sjogren’s syndrome etc.Therefore,novel drugs are required to supplement treatment modalities and to improve the prognosis of the advanced OSCC patients.Anlotinib is a domestic small-molecular tyrosine kinase inhibitor,targeting multiple angiogenetic factor receptors to suppress the angiogenesis and to inhibit partial functions of tumor cells.Multiple clinical researches have indicated the survival benefits of anlotinib in non-small cell lung cancer,hepatic cell carcinoma,medullary thyroid carcinoma and other malignances.However,few researches focus on the impact on OSCC.Given the role of tumor angiogenesis in OSCC and the working mechanism of anlotinib,it is prospective to research the impact of anlotinib on OSCC cells.Objectives: By detecting the impact of anlotinib on Tca8113 cell proliferation,invasion and migration functions to explored its impact on tongue squamous cell carcinoma cells in vitro,and to provide initially theoretical basis for clinical application of anlotinib in treating advanced OSCC patients.Methods: We cultured human tongue squamous carcinoma Tca8113 cells in vitro,and added cell culture medium containing 10 mmol / L anlotinib(drug group)and complete cell culture medium with the same dose(solvent control group).By performing CCK-8 assay,we detected absorbance value(OD)and compared the difference of cell proliferation between two groups.By performing transwell assay,we observed invading or migrating cells and compared the difference of cell migration or invasion between two groups.By wound healing assay,we observed the healing of the scratch area and compared the differences of cells migration between the two groups.Results: CCK-8 assay showed that the OD value of anlotinib group was significantly reduced than that of control group(P<0.05),which indicating that the proliferation of Tca8113 cells in anlotinib was significantly inhibited.Transwell assay showed the number of cells in anlotinib group invading through Matrigel medium was significantly reduced,average 39 cells in anlotinib group and 133 cells in control group(P<0.05).And the number of cells in anlotinib group immigrating to the bottom of chamber was also significantly reduced,average 161 cells in anlotinib group and 318 cells in control group(P<0.05).Wound healing assay showed that anlotinib significantly inhibited Tca8113 cells migrating to the scratch area(P<0.05),and the wound healing rate in anlotinib group was also reduced(19.81% VS 27.88%,P<0.05).Conclusion: Anlotinib significantly inhibited Tca8113 cells proliferation,invasion and immigration,which indicating the inhibition on tongue squamous cell carcinoma cells in vitro.However,researches are required to explore its inhibition in vivo,clinical activity and safety etc.,before it becomes a novel treatment alternative for advanced OSCC patients.
